【摘要】 目的:研究蟾蜍灵(bufalin)对人急性淋巴白血病CEM细胞的作用及机制。方法:台盼蓝拒染法观察蟾蜍灵对细胞的增殖抑制作用,Wright—Giemsa染色观察细胞凋亡形态学改变,流式细胞仪分析检测凋亡。结果:(1)10nM以上蟾蜍灵作用24h以上明显抑制CEM细胞生长(p<0．01), 且蟾蜍灵以时间、剂量依赖方式抑制CEM细胞增殖,24h、48h、72h半数抑制浓度(IC50)分别约为15．13nM、 9．59nM、6．57nM;(2)20nM以上浓度蟾蜍灵作用CEM细胞24小时出现典型细胞凋亡形态学改变,表现为细胞核固缩、碎裂、细胞膜完整并保持台盼蓝拒染功能及凋亡小体:(3)20nM蟾蜍灵处理CEM细胞12h、24h、48h,凋亡百分率分别为2．46％、17．92％、66．26％,40nM蟾蜍灵处理CEM细胞12h、24h、48h引起的凋亡率分别为5．19％、32．71％、80％以上,凋亡率与蟾蜍灵浓度呈剂量和时间依赖关系。结论:蟾蜍灵能够诱导急性淋巴白血病CEM细胞凋亡,其对CEM细胞的增殖抑制作用是通过凋亡实现的。更多还原

【Abstract】 Objective: To investigate the mechanism of inhibiting actions of bufalin on human acute lymphoblastic cell line CEM. Methods: Growth inhibition by bufalin was analyzed by trypan-blue staining. Apoptotic cells were detected by morphology and flow cytometric analysis . Results: l.Cell viability was significantly decreased by 10nM bufalin treatment after 24h,(p<0.01), the viability was significantly decreased by bufalin in time- and dose-dependent manner .The IC 50 (24h,48h,72h) of bufalin was about 15.13 nM, 9.59 nM ,6.57 nM,. 2. Apoptosis changes of morphology was observed by 20nM of bufalin or higher (24h),including cell shrinkage,chromatin agglutination and formation of apoptotic corpuscular.3 .The apoptosis rate of CEM cells treated by 20nM bufalin (12h,24h,48h) was 2.46%,17.92%,66.26%, the apoptosis rate of CEM cells treated by 40nM bufalin (12h,24h,48h) was 5.19%,32.71%, more than 80%, the apoptosis rate was significantly increased by bufalin in time- and dose-dependent manner. Conclusion:Bufalin can effectively induce apoptosis of CEM cells,which is possibly one of the mechanisms for inhibiting actions of bufalin in CEM cells.更多还原
