春砂仁多糖的分离纯化及分析

【摘要】 本文首次报道春砂仁多糖的分离纯化及结构分析的研究。春砂仁粉经超声波水提醇沉,并除去其中的蛋白质,得到春砂仁多糖。以凝胶过滤色谱法测定多糖的数量及分子量,结果为:春砂仁主要含两种多糖,其数均分子量分别为56421及1743,经分析,推出两种春砂仁多糖中单糖单位分别为329个及10个。多糖经酸解成为单糖并衍生为乙酰化的糖肟,对照标样采用气相色谱法测定其单糖组分为:阿拉伯糖、甘露糖、葡萄糖及半乳糖,四种单糖含量的相对比例为:1.000:0.681:0.972:0.862。
【Abstract】 The paper reported for the first time the purification and analysis of the Amomum villosum Lour. polysaccharides. In the test, the dry powder of Amomum villosum Lour. was extracted with ultrasonic method by water and deposited by ethanol, then the protein in it was removed by Sevag method., which resulted in the polysaccharides. The number and molecular weight of them were tested by Gel Filtration Chromatography(GFC), which showed that there are two kinds of polysaccharides in Amomum villosum Lour. and that their Mn are 56421 and 1743, respectively. After the polysaccharides were hydrolyzed in acidic condition, the corresponding monosaccharides formed and were transformed into acetyl derivatives by acetic anhydride. The derivatives were tested by Gas chromatography (GC) to give the result that the four monosaccharides exist with the certain proportion : arabinose : mannose : glucose : galactose = 1.000:0.681:0.972:0.862. By analysis, the number of the monosaccharides in the two kinds of the Amomum villosum Lour. polysaccharides is probably 329 and 10.
