基于SiO2溶胶-凝胶的电化学免疫传感器的研究

【摘要】 结合SiO2溶胶-凝胶(Sol-gel)技术,以石墨电极为基础电极,利用SiO2溶胶-凝胶的吸附力强、生物亲和性大的性质将辣根过氧化物酶吸附到电极的溶胶-凝胶表面,以对苯二酚为电子媒介物制备酶传感器用于测定H2O2,在最大H2O2浓度响应下,用SiO2溶胶-凝胶结合抗体, 抗体再结合抗原,并将其用于固定含二抗的辣根过氧化物酶,以对苯二酚为电子媒介物制得用于测定免疫球蛋白抗原的电化学免疫传感器。采用循环伏安法对传感器逐层修饰过程进行了表征,讨论了溶液pH值、抗体稀释度对电极性能的影响。结果表明,该方法能保持被固定蛋白质分子的活性,增强了传感器的稳定性;该传感器在2.5×10-5-12.5×10-5g·L-1免疫球蛋白抗原浓度范围内与电流响应值呈良好的线性关系,检测限为2.0×10-5g·L-1,且响应迅速、灵敏度高、选择性好,在实际应用中结果良好。

【Abstract】 With the graphite electrode as foundation electrode, a new type of electrochemistry immunesen-sor for the determination of H2O2 was made by combining sol-gel techniques and to link antibody(Ab) on the sol-gel film surface, so the antigen was immobilized on the antibody, horseradish peroxidase ( HRP) was immobilized . The sol-gel/Ab/Ag/HRP layer was immobilized on the surface of the graphite electrode. Cyclic voltammetry was employed to investigate the structure and the electrochemical characteristics of the immun- nosensor.The influence factors related to the sensitivity of the system such as pH , the titer of antibody. The experimental results show that the activity of the protein is maintained better by this method, and the stability of the immunosensor is improved . under the biggest H2O2 response, and the enzyme electrode dis-playes an excellent electrocatalytical response linearly to the reduction of antigen in the concentration range of 2.5×10-5-12.5×10-5g·L-1. The detection limit is 2×10-5 g·L-1.
