用SSR标记研究核桃野生群体的遗传多态性

【摘要】 利用简单重复序列(SSR)对我中国核桃两主要栽培种-核桃(J。regia L.)和铁核桃(J.sigillate Dode)的8个群体(共240个体)的遗传多态性、种群结构及遗传分化进行分析。结果显示,8对SSR引物共检测到71条等位基因,平均每个位点8.88条。其中铁核桃群体61条,核桃群体49条。多态位点比率在两个种中都为100%。遗传多态性分析结果显示,铁核桃群体的平均期望杂合度(He)为0.5886, 大于核桃群体的(He)0.5213,铁核桃与核桃两个种群体间的平均分化系数Fst值分别为0.0729和 0.1349,说明只有7.29%的变异存在于铁核桃群体间,而有13.49%存在于核桃群体间。而铁核桃与核桃种两个种间的分化系数Fst值为0.1034。数据表明分布在西南地区的铁核桃群体及核桃群体具有较为丰富的遗传多态性,但由于人为干扰,群体间分化程度降低,造成稀有基因资源流失,因此应尽快对该地区的核桃资源实施保护,以及进行合理利用。对于铁核桃与核桃两个种之间的亲缘关系,还有待进一步研究。
【Abstract】 The genetic structure and diversity of 8 semi-nature population (240 individuals total) of two main cultivated species of J. sigillate Dode and J. regia L. were investigated based on 8 microsatellite loci. a moderate to high level of genetic diversity was observed at population levels with the total number of alleles 71 and average 8.88 for per locus, 61 of them belong to populations of J. sigillate Dode and 49 belong to populations of J. regia L.The proportion of polymorphic loci (P) equaling 100.0%. The expected heterozy-gosity (He) in populations of J.sigillate Dode with an average of 0.5886 which is higher than that (0.5213) in populations of J. regia L., The proportion of genetic differentiation present among populations accounted for 7.29% of the whole diversity in populations of J. sigillate Dode, 13.49% in populations of J. regia L., All the statistic indicates that the populations distribute in southwest of China appear higher genetic diversity than other populations,but for the disturbances of human being, the differences between populations is declining gradually and then will cause the disappearance of rare gene. So we should carry out strategy of protection and management as soon as possible. In addition, the relayionship between the two species is still in argument at present.
