大豆与豆科模式植物蒺藜苜蓿的穿梭标记研究

【摘要】 利用生物信息学和比较基因组学的研究方法,基于已知的大豆RGA序列,设计了36对SSR 引物(从与已知的RGA序列具有相似性的蒺藜苜蓿BAC中搜索SSR标记设计引物),对大豆的4个遗传群体(03169、03197、03172、Jinf)进行多态性筛选,研究结果显示,Smt5021标记在03197群体上具有多态性,在F2群体表现出良好的分离模式,Smt4731标记在03169群体上有多态性,在F2群体中出现明显的偏分离现象;Smt1332标记在03169、03197以及03172三个群体上均有多态性。利用JoinMap方法将Smt5021标记定位在G2连锁群上,与两个相邻的大豆SSR标记距离分别为3.5cM(Satt303)和6.7cM (Satt131),而与两个相邻的AFLP标记距离分别为0.7cM(H1-7)和1.5cM (H24-16);初步研究表明, Smt5021标记是一个理想的大豆与蒺藜苜蓿间的穿梭标记(cross species marker)。
【Abstract】 Based on the utilization of bioinformatics and comparative genomics, 36 SSR primer sets were designed according to soybean RGA sequence, i.e. screened those SSR markers with analogous sequence to RGA sequence in the BAC of Medicago truncatula Gaertn. Subsequently, the four soybean genetic population, i.e. No.03169, No. 031970, No.03172 and Jinf, were sceened using these SSR markers. The results indicated that Smt5021 showed polymorphism in the populaton No. 03179, and segregated in a good Mendel model. Smt4731 showed polymorphism in the populaton No. 03169 and segretated in a bias model. Smtl332 showed polymorphism in the populaton No. 03169, No. 03197 and No. 03172. The marker Smt5021 is located on linkage group G2, which is 3.5cM (Satt303) and 6.7cM (Satt131) away from the two neighbor markers, and 0.7cM (H1-7) and 1.5cM (H24-16) away from the two AFLP markers. The results indicated that Smt5021 was an ideal cross-species marker between soybean and Medicago truncatula Gaertn.
