一个内含子长度多态性标记与栽培稻F1花粉育性基因座连锁

【摘要】 本研究利用两份栽培稻(Oryza sativa L.)种质HITAR005和IRGC20509杂交建立了含有500 个单株的F2群体,采用内含子长度多态性标记对F2群体中的117株进行了标记基因型分析。研究发现一个内含子长度多态性标记,RI01594,其标记座位上与父本(IRGC20509)相同基因型的纯合植株完全消失,且母本纯合基因型植株与杂合基因型植株的比率符合1:1(x2c=0.90,X2c<X20.05),表现出一种明显的偏分离现象。分析该分子标记所在的目的序列,RI01954标记所位于的基因含有3个内含子是一个功能未知的转录因子,粳稻Nipponbare(日本晴)在该基因的第3个内含子序列与籼稻9311的序列相比有24个碱基的缺失。进一步分析RI01954标记的PCR产物表明,发现消失的纯合植株基因型偏向于粳稻Nipponbarc(日本晴)。结合供试的F2群体的花粉育性检测结果,初步表明RI01594标记与栽培稻F1花粉育性基因座连锁,与F1花粉不育基因座连锁的遗传距离小于0.5cM,推测与RI01954标记连锁的栽培稻的F1花粉不育性基因座是一个新的杂种不育位点。

【Abstract】 The F2 population containing 500 individuals was developed using two cultivated rice germplas-ms (Oryza sativa L.), HTTAR005 as female parent from HTTAR and IRGC20509 as male parent from IRRI, and the ILP (intron length polymorphism) markers were employed to genotype the 117 individuals of the F2 population. RI01954, an BLP marker mapped in the 3ri chromosome, was found that its homozygous locus for male parent were completely absent in the 117 individuals and the ratio of RI01954 locus between homozygous of female parent and heterozygous was 1:1 (x2c=0.90, x2c<X20.05), which showed obviously distorted segregation in the F2 population. The sequence conferring RI01954 was deeply analyzed that it is unknown functional transcript factor containing three introns. There is 24bp deletion of the 3rd intron in Japoni-ca Nipponbare compared with Indica 9311. Analysis of PCR products amplified by primers of RI01954 showed that the disappeared bands of male parent were identical to Japonica Nipponbare. Combining the result of pollen fertility analysis in the F2 population, it was preliminarily concluded that RI01954 marker closely linked to the phenotype of the F1 pollen sterility locus and the genetic distances between them was estimated less than 0.5cM. And also it suggested that the locus linked with RI01954 was a novel F, pollen sterile locus in the cultivated rice.
