HPLC法测定九味羌活颗粒中欧前胡素、异欧前胡素的含量

【摘要】 目的:建立九味羌活颗粒中有效成分欧前胡素、异欧前胡素的含量测定方法。方法: 样品进行处理时,对提取香豆素类成分所用的溶剂种类,极性和提取方式进行了系统研究, 最后选用乙酸乙酯为最佳溶剂,提取九味羌活颗粒有效成分可获得较高的提取效率。用乙酸乙酯超声处理,静置过滤,于65-70℃水浴蒸干,最后用甲醇定容,高效液相色谱法测定九味羌活颗粒的欧前胡素、异欧前胡素的含量。色谱柱:C18柱(汉邦科技,200mm×4.6mm, 5 m),乙腈-水(55:65,V/V)为流动相,流速为1.00ml/min,检测波长为250nm。结果: 欧前胡素在0.180-1.200 g范围内、异欧前胡素在0.120-1.200 g线性关系良好,表明此法可作为九味羌活颗粒中欧前胡素、异欧前胡素的含量测定,用于九味羌活颗粒质量标准的制定。结论:所建立的方法简便可行,能准确、快速地进行九味羌活颗粒中欧前胡素、异欧前胡素的定量分析与检测:重复性较好,可用于该制剂的质量控制方法。
【Abstract】 Objective:The essay studied granule preparation of Jiuweiqianghuo Granule and established an HPLC method for determination of Imperatorin and Isoimperatorin in Jiuweiqianghuo Granule. Method:When disposed of sample , the solvent varieties, polarities and extraction methods were systematically studied . At last, acetoacetate was choosed as the better solvent , because of the more way to extract Imperatorin and Isoimperatorin from Jiuweiqianghuo Granule . The sample was extracted by ultrasonic wave in acetoacetate, standed and filtered, evaporated to dryness on a water bath (65~70℃).Then methnol made it to 10ml. The HPLC method was carried out on C18(HanBang 200nm×4. 6nm, 5 mcolumn with acetonitrile-water solution as a mobile phase .The flow rate was 1. 00ml/min.The detection wavelength was at 250nm and the column temperature was 30℃. Reasult:The standard cure was linear within the concentration range of 0.180~1. 20 g Isoimperatorin and of 0.120~1. 200 g Imperatorin, which indicated the method can be used to the determination of Imperatorin and Isoimperatorin from Jiuweiqianghuo Granule and the formulation of Jiuweiqianghuo Granule. Conclusion:The established method was simple, workable , accurate, rapid and reproducible .It could be used for quality control for Jiuweiqianghuo Granule.as a commodity.
