MBP特异性T细胞系的建立(英文)

【摘要】 目的建立MBP特异性自身反应性T细胞克隆,为进一步探讨EAE的发病机制和治疗方案奠定基础。方法以MBP肽免疫C57BL/6小鼠,取其淋巴结细胞,加同系鼠脾细胞体外共同培养于含IL-2的培养液中,并以 MBP肽反复刺激。收集培养细胞,以流式细胞仪进行鉴定。结果流式细胞仪分析显示,90%以上的细胞为CD4+ T细胞。结论成功地建立了C57BL/6小鼠的MBP反应性T细胞克隆,该细胞克隆为MBP特异性自身反应性 CD4+T细胞。
【Abstract】 Objective To establish myelin basic protein (MBP) autoreactive T cell line for investigating the pathogen mechanism and treatment method of EAE. Methods Lymphocytes were taken from lymph nodes of C57BL/6 mice immunizd with MBP peptide 68 ~ 86 and cultured in. vitro with syngenic splenocytes in medium which LI-2 and MBP p68 ~ 86 was added into. Some days later, the cells were collected and identified with FACS. Results The result showed that more than 90% of the cells were MBP-specific CD4+ T cells. Conclusion MBP-specific T cells line which belonged to CD4+ subest is successfully established from C57BL/6 mice.
