组织工程学构建兔关节软骨细胞-羊膜支架复合体

【摘要】 目的:研究羊膜作为透明软骨细胞组织工程支架材料的可行性。方法:取胎兔膝关节软骨细胞进行体外原代培养与传代培养,把第二代软骨细胞接种到羊膜上,进行光镜、扫描电镜观察。结果:(1)关节软骨细胞于羊膜上牢固粘附,6天开始增殖,3周时基本铺满羊膜表面;(2)去掉上皮细胞的羊膜细胞吸附性、体外生物相容性强于未去掉上皮细胞的羊膜。结论:羊膜表现良好的细胞吸附性和生物相容性,有可能成为组织工程中一种良好的细胞培养支架。
【Abstract】 Objective: To study the possibility of the amnion as hyaline cartilage cells scaffold in tissue - engineering. Method: To carry the rabbit’s knee joint hyaline cartilage cells on original culture and spread culture outside body, inoculate the second generation cartilage cell to the amnion, and observe the composite in the light mi-croscop, scanning electronic microscopy. Results: The joint hyaline cartilage cells attached to the amnion closely and begin proliferate after 6 days , spreaded mostly the full amnion surface 3 weeks later. Conclusion: Amnion have good adsorptivety and biocompatibility with hyaline cartilage cells, it can be used as scaffold for tissue - engineering.
