芦笋AFLP技术体系的建立与优化

【摘要】 以本课题组自行培育的芦笋品种A10为试材,通过对反应体系中的几个关键因素进行优化,建立了适宜芦笋的AFLP体系。结果表明:酶切基因组适宜用量为400～500ng; 酶切-连接采用一步法;预扩增时退火温度降低5℃;预扩增产物适宜稀释倍数为20倍。更多还原

【Abstract】 An AFLP ( amplified fragment length polymorphism) analysis system was established with Asparagus A10 which were bred by our research group by optimizing factor of the AFLP technological system. The result showed that the dosage of template DNA should be 400 -500ng; the digestion and ligation should be done in one step, and the products of pream-plification should be diluted to 20 times.更多还原
