几种壳聚糖改性材料蛋白吸附作用比较的实验研究

【摘要】 [目的]对三种改性壳聚糖生物材料进行蛋白吸附作用的比较实验研究。[方法]应用MTT比色法,测定BSA-CGG,CGG,CG三种材料对大鼠原代表皮细胞的细胞毒性和细胞增殖率;同时应用考马斯亮兰G-250染色法测定材料的蛋白吸附作用。[结果]本实验结果显示,壳聚糖材料的细胞毒性结果为阴性,不同壳聚糖材料不同时间对蛋白的吸附量有差异,同种材料不同时间对蛋白的吸附量差异不大。[结论]壳聚糖改性材料在细胞体外增殖作用和蛋白吸附作用方面要优于壳聚糖。
【Abstract】 Objective Investigate the cytotoxicity and protein adsorption of dimensional porous chitosan scaffolds. Methods The keratinocytes of rat were seeded with the BSA-CGG,CGG,CG respectively, the cytotoxicity was detected by MTT. The protein adsorption of modified scaffolds (hyaluronate-chitosan scaffold and hyaluronate-BSA-chitosan scaffold) were detected by G-250. Results The modified chitosan scaffolds show no cytotoxicity. Two kinds of modified chitosan scaffolds adsorbing protein were different and the same modified chitosan scaffolds adsorbing protein were different in 2h, 4h, 8h, 24h respectively.. Conclusion The modified chitosan scaffolds have satisfactory biocompatibility and adsorbs protein very well.
