三七总皂甙经细胞外信号调节激酶信号通路诱导血管平滑肌细胞增殖
Total Panax Notoginseng Saponin Induces Proliferation of Cultured Vascular Smooth Muscle Cells Mediated by ERK Signal Pathway

【摘要】 目的血栓通是从中药三七中提取有效成份三七总皂甙而制成。研究发现三七总皂甙具有抑制和促进细胞增殖的双向调节作用。但它通过什么机制导致双重作用仍然不清。本研究旨在观察三七总皂甙是否可诱导血管平滑肌细胞增殖,并经由什么信号通路介导。方法用分子生物学、生物化学方法观察体外培养的血管平滑肌细胞在一定的三七总皂甙浓度作用下,血管平滑肌细胞的细胞外信号调节激酶磷酸化及细胞增殖的变化。结果三七总皂甙能快速地激活ERK1/2,呈时间依赖性。在三七总皂甙浓度为0.2g/L 作用8min 时,引起明显地细胞外信号调节激酶磷酸化,呈现最大程度的激活,随后逐渐减弱,4h 时恢复到基线水平。同时,三七总皂甙也可引起血管平滑肌细胞增殖增加。在三七总皂甙浓度为0.1g/L 时,三七总皂甙能引起细胞增殖。在浓度为0.2g/L 时,细胞增殖数达最大,无三七总皂甙作用组(阴性对照组)与各三七总皂甙组之间差异均有显著性。然而,随着三七总皂甙浓度的增加,三七总皂甙引起血管平滑肌细胞增殖的效应渐弱。结论三七总皂甙经由细胞外信号调节激酶磷酸化途径是引起细胞增殖的机制之一。
【Abstract】 Aim It has been showed that total panax notoginseng saponin(TPNS) has dual effects on proliferation andinhibition of cells.However,the mechanism by which TPNS leads to dual effects is unclear.To explore whether the TPNS in-duces VSMC proliferation and by which signal pathway mediates.Methods Using approaches of molecular biology,biochem-istry,the changed extracellular signal-regulated kinases (ERK1/2) phosphorylation and VSMC proliferation were observed.Results VSMC treated with TPNS (0.2 g/L) resulted in a time-dependent induction of ERK1/2 phosphorylation that peaked at 8min and declined thereafter and returned to basal line by 4 h.Meanwhile,’rPNS also induced VSMC proliferation that was evi-dent at 0.1 g/L TPNS,and peaked at 0.2 g/L and then declined with increase of TPNS concentration.The effects of TPNS onVSMC proliferation was significant difference compared to negative control (without TPNS treatment).Conclusion Activationof ERK1/2 signal pathway in response to TPNS in vitro leads to increased VSMC proliferation.
