雷公藤蒽醌成分大黄素对小鼠迟发型超敏反应及脾淋巴细胞增殖反应的影响

【摘要】 目的观察体外具有抑制T淋巴细胞增殖活性的雷公藤蒽醌成分大黄素是否在体内具有免疫抑制活性作用。方法腹腔给药,观察大黄素对二硝基氟苯(DNFB)诱导的小鼠迟发型超敏反应的影响。同时,采用~3H-甲基胸腺嘧啶核苷(~3H-TdR)掺入法,观察大黄素体内给药后小鼠脾淋巴细胞对Con A诱导的T细胞以及脂多糖(LPS)诱导的B细胞增殖反应中的影响。结果大黄素腹腔给药对二硝基氟苯诱导的小鼠迟发型超敏反应有显著抑制作用,给药后小鼠脾淋巴细胞对丝裂原诱导的增殖反应能力明显低下。结论大黄素具有较强的免疫抑制活性。

【Abstract】 Objective To observe whether emodin, an anthraquinone component in Tripterygium wilfordii Hook.f. which can inhibit the proliferation of T lymphocyte in vitro, has the immunosuppressive activitier in vivo. Methods To observe the effect of emodin on delayed type hypersensitivity (DTH) induced by dinitrofluorobenzene (DNFB), mice were administered emodin by i.p. In the meantime, spleen cells from mice having been injected emodin were stimulated by concannavalin A(Con A) and lipopolysaccharide (LPS) respectively,and proliferation of T and B lymphocytes were measured with ~3H-methylthymidine (3H-TdR). Results Emodin had outstanding inhibition on DNFB induced DTH response in vivo(i.p.); T and B lymphocytes from emodin trcatted mice showed the low protifertive repronse to Con A and LPS stimulation. Conclusions Emodin has significant immunosuppressive action in vivo.
