高效液相色谱-蒸发光散射检测法测定烟草中的水溶性糖

【摘要】 以乙腈/水=70/30作为流动相,采用蒸发光散射检测器,建立了高效液相色谱同时测定烟草中鼠李糖、木糖、阿拉伯糖、果糖、甘露糖、葡萄糖、蔗糖、麦芽糖等8中水溶性糖的新方法。上述8种糖的加标回收率分别为:96.6%、92.2%、86.0%、102.4%、90.8%、101.2%、98.6%、97.5%。线性范围分别为:鼠李糖、果糖、葡萄糖、蔗糖0.4~20μg:木糖、阿拉伯糖、甘露糖、麦芽糖0.8~20μg。相对标准偏差皆小于4.1%。测定不同烟叶及卷烟烟丝中的水溶性糖。方法准确,结果稳定,重现性好,可用于烟草及其制品中糖含量的质量分析与控制。

【Abstract】 A new approach for the determination of 8 carbohydrates (rhamnose, xylose, arobinose, fructose, mannose, glucose, sucrose, maltose) in tobacco extract was described. The analytical method was based on the extraction of carbohydrates from tobacco samples with a water solution, followed by isolation of target analytes from extract aliquots by solid-phase extraction using Sep-pak-C18 cartridges. The extracts were analysed by liquid chromatography with evaporative light scattering detector. The 8 carbohydrates were separated on high performance carbohydrate column using an acetonitrile-water(70:30, v/v) at a flow-rate of 1.0 mL/min. The recoveries from spiked extracts were high and reproducible, ranging from 86.0~102.4%,at concentration levels that were as expected following extraction from average quality tobacco material. The method had been successfully applied to determine the individual carbohydrates in tobacco leaves and tobacco cut.
