中药复方TCMP-1对急性胰腺炎血小板内皮细胞粘附分子-1表达变化的研究

【摘要】 <正>目的;探讨急性水肿性胰腺炎(AEP)血(?)内皮细胞粘附分子-1(PECAM-1,CD31)表达的变化规律及中药复方TCMP-1对AEP的治疗作用。方法:用皮下注射蛙皮缩胆囊肽的方法诱发大鼠AEP动物模型,经脾静脉和下腔静脉采血,用流式细胞仪分析体循环和微循环白细胞PECAM-1的表达。

【Abstract】 Objective To investigate the changes of platelet endothelial cell adhesion molecule-1(PECAM-1) expression in leukocyes in acute edematous pancreatitis (AEP) and the effeces of Chinese medicine on AEP. Method The model of AP was established by subcutaneous injection of cacrulein, the changes of expression of PECAM-1 in leukocytes from splenic vein and inferior vena cava was determined by flow cytometry. Result (1) To compare with control group, all rats treated with caerulein showed hyperamyleimia (P<0.05)cxcept the AEP8h group (P>.05). (2) To compare with control group, the dry/wet pancreatic weight ratio decreased graduallyin all experimental groups, and the reduction is significant in all the acute edematous pancreatitis(AEP) group(P<0.05), but not in the TCMP-1 treated group. (3)In the model of the AEP group, PECAM-1 expression in polymorphonuclear leukocytes (PMN) was close to each other in AEP2h and AEP4h group. Since AEP4h group, PECAM-1 expression in pancreatic microcirculation was down-regulated till AEP8h group; PECAM-1 expression in systemic circulation was up-regulated till AEP8h group, and the difference of PECAM-1 expression was significant between microcirculation and systemic circulation in AEP8h group(P<0.05). (4) In the model of the TCMP-1 treated group, to compare with the AEP group, PECAM-1 expression in PMN in pancreatic microcirculation and systemic circulation was reduced in different degrees and the reduction is significant in systemic circulation in the AEP8h group(P<0.05). Conclusion up-regulation of PMN PECAM-1 expression systemic circulation may regulate the affinity of integrin in helping PMN to adhere to endothelial cell firmly and to transmigrate through endothelium; down-regulation of PMN PECAM-1 expression in microcirculation is related to the activation of PMN and can facilitate PMN transmigrating. Thus the TCMP-1 may prevent PMN from transmigrating through endothelium by the down-regulation of the basic level of PMN PECAM-1 expression or the inhibition of overexpression of PMN PECAM-1.
