应用PAS-ELISA和RT-PCR检测柑橘衰退病毒

【摘要】 本研究采用PAS-ELISA法和3种RT-PCR方法对柑橘衰退病毒(CTV)进行了检测。结果表明,在3~7月份采集的柑橘样品PAS-ELISA法可以准确地检测出CTV,茎尖、叶柄和韧皮部组织病毒浓度较高,是较理想的检测材料。RT-PCR、IC-RT-PCR和TC-RT-PCR这3种方法在病毒浓度较低的季节,仍能得到理想的检测效果,其中IC-RT-PCR更适合用于病毒浓度较低时的检测。

【Abstract】 In this study, PAS-ELISA and three different RT-PCR methods were used to detect Citrus tristeza virus (CTV) in citrus tissues. The results showed that ELISA could reliably detect CTV in citrus samples collected from March to July and shoot tips, petioles and barks were idealistic tissues for CTV detection. Three molecular methods including RT-PCR、IC-RT-PCR and TC-RT-PC could give definitively positive results for CTV in samples collected during growing seasons. Among these, IC-RT-PCR was more suitable for CTV detection during seasons with lower virus content.
