超声波空化效应对体外培养细胞细胞膜作用的实验研究

【摘要】 目的研究超声空化作用对体外培养细胞细胞膜的生物学效应,探讨声学造影剂增强基因转染的机制。方法将血管平滑肌细胞悬液分为对照组、单纯超声辐照组和造影剂+超声辐照组三组,分别给予频率为665kHz、声强为0.5W/cm2的超声波辐照20s,实验后立即和继续培养24小时后用扫描电镜观察细胞膜结构有无改变。结果单纯超声辐照组细胞及细胞膜的结构与对照组无显著差异。造影剂+超声辐照组约20%的细胞膜表面出现了大小约1~2μm左右的小孔。24小时后再次观察细胞膜表面恢复正常。结论空化效应使细胞膜上出现可逆性小孔。致细胞膜的通透性增高可能是声学造影剂增强基因转染的主要机制。

【Abstract】 Objective To investigate the effects of ultrasonic cavitation on cell membrane cultured in vitro. Methods The rat pulmonary artery vascular smooth muscle cells were divided into three groups, the first group was control group, the second and third group were exposed to ultrasound(655kHz, 0. 5W/cm2) for 20s with or without contrast agents respectively. Immediately and 24 hours after treatments, electron microscopic scanning were performed. Results Compared with control group, the structure of cell and cell surface had no apparent changes in the second group treated with ultrasound alone. In the third group, nearly 20% cells exhibited small holes with diameter mostly about 1-2μm in the cell surface and returned to a normal appearance after 24 hours later. Conclusion Ultrasonic cavitation can cause small transient holes in cell surface and hence increase permeability of the cell membrane. It might be the main mechanism for gene delivery with ultrasound contrast agents.
