哺乳动物克隆再编程中的表遗传机制

【摘要】 虽然利用核移植技术已经成功地克隆了多种哺乳动物,但是此过程仍存在很多问题,例如孕早期高流产率和出生前死亡等。导致这些克隆动物发育失败的一个因素在于供体体细胞基因组不完全表遗传再编程所致遗传修饰出现偏差和失误,尤其是去甲基化异常的出现。我们探讨了哺乳动物早期克隆胚胎再编程机理,期望在解决克隆技术的问题能提供一些线索。

【Abstract】 Despite the fact that cloned animals derived from somatic cells have been successfully generated in a variety of mammalian species, there are still many unsolved problems in current cloning technology. Nuclear cloning has shown some developmental aberrancies, including a high rate of abortion during early gestation and increased prenatal death. One of these failures of cloned embryos may due to the epigenetic reprogramming of somatic donor genome. Some results suggest that the developmental failures of cloned embryos may be associated with the errors and abnormalities of epigenetic reprogramming of donor genome DNA. Understanding molecular mechanisms for reprogramming of donor genome will contribute to clarify the developmental defects of cloned embryos and aid improvement in biotechnologies applied to early embryos in all species.
