晋南牛5个微卫星位点多态性的研究

【摘要】 利用PCR技术和电泳银染技术检测了晋南牛TGLA227,ETH225,HEL9,CSSM66,TGLA1216等5个微卫星位点的多态性。结果表明,5个微卫星位点均处于不平衡状态(P<0.01),其中TGLA126位点H,Ne,PIC最高,分别为0.9765,0.8858,8.9407。5个微卫星基因座的平均杂合度为0.9809,平均多态信息含量为0.9049,平均有效等位基因数为10.1636,这些结果都表明晋南牛具有丰富的遗传多样性。

【Abstract】 The polymorphism of five microsatellite loci,TGLA227, ETH225, TGLA126, HEL9, CSSM66, were detected in Jinnan cattle by polymerase chain reaction and 8% PAGE electrophoresis followed by silver staining. Allelic frequency (Pi), heterozygosity(H), polymorphism information cotent (PIC) were caculated. Five loci were not at Hardy-Weinberg equilibrium(P<0.01), HJPIC and Ne of TGLA126 loci are the biggest among five loci, Average H of five loci is 0.9809, Average PIC of five loci is 0.9050, Average Ne of five loci is 10.1636, These result showed five microsatellite loci detected were all polymorphic in Jinnan cattle, which could be used for linkage analysis, individual identification and paternity test in Jinnan cattle.
