更年宁心胶囊与戊酸雌二醇对更年大鼠脾细胞凋亡基因Bcl-2、Bax表达的影响

【摘要】 目的:比较更年宁心胶囊与戊酸雌二醇对更年期大鼠脾细胞凋亡基因Bcl-2、Bax 的表达。方法:30只12月龄SD雌性大鼠随机分为3组:更年组(每日予蒸馏水灌胃1ml/只),雌激素组(戊酸雌二醇800μg.kg-1.d-1灌胃),中药组(更年宁心胶囊2.5g.kg-1.d-1灌胃)。3个月后颈动脉放血处死。放射免疫法测定血清E2水平,Annexin V-FITC/PI双染色流式细胞仪测定脾细胞凋亡率,RT-PCR技术检测脾细胞凋亡基因Bcl-2、Bax的表达。结果:雌激素组血清E2水平明显高于中药组与更年组,而脾细胞抗凋亡基因Bcl-2表达明显低于中药组与更年组。结论:与雌激素组比较,中药组增加了脾细胞抗凋亡基因Bcl-2表达,提示对淋巴细胞凋亡可能具有保护作用。凋亡相关基因的表达受到体内雌二醇浓度的影响。
【Abstract】 Objective: To compare Gengnianningxing with Estradiol Valerate on apoptosis-associated gene expressions in splenocytes of menopausal rats. Methods: Thirty 12 months old femal SD rats were randomly assigned to the following conditions: menopausal group, with distilled water 1ml/d po; estrogen group: Estradiol Valerate 800靏.kg-1.d-1 po; chinese herb group: Capsule of Gengnianningxing 2.5g.kg-1.d-1 po, After 3 months all rats were sacrificed by carotid artery exsanguination, Serum E2 levels were measured by radio-immunoassay. The apoptosis in splenocyte of rats was measurerd by Annexin V-FITC/PI flow cytometeic assay. The apoptosis-associated gene expressions were detected by RT-PCR procedures,.Results: The serum E2 level of estrogen group was higher than that of chinese herb group and menopausal group. but Bcl-2 gene expression was lower than that of chinese herb group and menopausal group. Conclusion: Compairing with ERT,Chinese herb group increased Bcl-2 gene expression in splenocytes,which was regulated by estrodiol concentrations.
