西瓜转葡聚糖酶及几丁质酶复合基因研究

【摘要】 采用根癌农杆菌(Agrobacterium tumefaciens)介导法将葡聚糖酶和几丁质酶基因共同导入西瓜(Citrullus lanatua)子叶外植体,经抗性筛选和组织培养获得再生植株。通过对转基因植株中插入序列的GUS染色鉴定和特异引物PCR检测。表明葡聚糖酶和几丁质酶基因已成功导入西瓜植株.
【Abstract】 A construct containing a rice chitinase gene and an alfalfa glucanase gene was co-transferred into watermelon through Agrobacterium tumefaciens-mediated method. Plantlets were regenerated in vitro by resistance selection on MS medium containing various concentions of kanamycin. GUS and PCR-aided assays indicated that both chitinase and glucanase genes have been co-transferred into watermelon successfully.
