榨菜细胞质雄性不育系及其保持系atpA基因组结构差异

【摘要】 以榨菜细胞质雄性不育系及其保持系基因组总DNA为模板,通过特异引物PCR方法克隆了榨菜atpA基因,基因大小为1524 bp,推定编码507个氨基酸,进化分析表明atpA基因非常保守;进一步分析发现了不育系及其保持系atpA基因的5’UTR区存在差异,这为能量代谢相关基因与细胞质雄性不育性产生的关系提供了分子证据。

【Abstract】 The purpose of this investigation is to compare the structural difference of atpA genes in cytoplasmic male sterile line and its maintainer line of tuber mustard. The intact atpA genes were cloned and determined to be 1524 bp in size,and deduced to code for 507 amino acids. Phylogenetic analysis indicated that it was highly conservative among Crucerfera family. However,there was obvious structural difference existent in upstream regulating region of atpA genes between CMS and M lines of tuber mustard. This finding will lay the basis for further clarification of the function of atpA genes in the involvement of cms expression.
