中国栽培大豆核心种质的取样策略

【摘要】 利用中国栽培大豆初选核心种质的农艺性状和60个SSR位点的分子数据,以9种不同方法,在9个取样水平上,分别构建核心种质,通过SSR等位变异保留比例、农艺性状表型符合度以及遗传多样性指数的比较,对大豆核心种质构建的最佳取样策略进行了研究。结果表明,分区法的代表性高于总体法;聚类取样法的代表性高于随机法;SSR数据聚类取样的SSR等位变异保留比例显著高于以农艺性状聚类取样,但农艺性状表型符合度低于以农艺性状聚类。此外,在区内取样量确定方面,平方根法的SSR水平代表性和遗传多样性shanno-weaver指数高于比例法,但农艺性状表型符合度和遗传多样性指数均低于比例法。因此确定以SSR分子指纹数据聚类,以平方根法确定组内取样量的分区法为构建中国栽培大豆核心种质的最佳取样方法。根据不同取样策略和不同总体取样量水平的核心种质遗传多样性指数比较发现,3%的取样水平不仅能够保证对初选核心种质较高的代表性,且核心种质具有较大的遗传多样性。

【Abstract】 9 methods and 9 sampling ratios, based on agronomic traits and. 60 SSR fingerprint, wereperformed to construct Chinese soybean core collection respectively The evaluation on the different corecollections suggest that core collection constructed by stratified by sub-regions have higher representativenessthan no stratification and selected by clustering is better than randomly selection; clustered by SSR data hashigher representativeness on SSR level, but lower representativeness on agronomic traits than clustered byagronomic traits. To determinate the amount of accessions in different subregions by Square root show higherrepresentativeness and diversity index than by proportion method on molecular level, on the contrary it islower than by proportion on phenotypic level. Considered several evaluation index, clustered by SSR data andselected by square root strategies were decided to be the optimal method for Chinese soybean core collection.According to the diversity index and representativeness, 3% was also decided as the optimal sampling ratiofor Chinese sovbean core collection.
