玉米类LFY基因的克隆及其在不同光周期条件下的表达

【摘要】 本实验采用每天9小时和15小时两种光照条件对热带玉米自交系CML288、温带玉米自交系黄早4和由它们组配的组合F1进行处理,观测调查其农艺性状的表现。结果表明:CML288对光周期的反应非常敏感,黄早4对光周期的反应不敏感。在玉米生长到3~10片时选取茎尖为材料。提取RNA,利用RT-PCR得到一个全长1265bp的cDNA克隆,序列分析发现该cDNA序列含有一个完整的1167bp的开放阅读框,编码一条约42kD的多肽,包含389个氨基酸。对氨基酸序列进行二级结构分析,结果显示其含有21个卷曲,17个螺旋,6个折叠股。表达分析结果表明,在长日照条件下,该基因在2个供试自交系不同生长时期的茎尖中持续表达;在短日照条件下,在2个自交系生长的早期和后期的茎尖中不表达。从其表达情况上看,该基因的功能可能不是促进开花,具体功能和作用的机理以及生理生化过程还有待进一步研究。
【Abstract】 Two inbreds, CML288 from CIMMYT and Huangzao 4 from temperate zone and their hybrid F1 were selected for study. Plants were grown in 2 different controlled photoperiod, 9hr and 15 hr. Growth stages, plant and ear height, leaf number were examined. The results showed that CML288 was very sensitive to photoperiod and could only grow in vegetative phase, not flower; but Huangzao 4 was on the contrary. Extracted RNA from the stern top of Maize we isolated a 1265bp cDNA clone from maize by RT-PCR. The clone contains an open reading frame encoding a putative protein of 389 amino acids. There were 21 coils, 17 helices and 6 strands in the secondary structure of the protein. Expression analysis showed the gene expressed all time under long day, while not early or late under short day. The function of the gene have been at least partly unknown and need more research.
