液相色谱-质谱联用测定人体血浆中的非那雄胺

【摘要】 <正>非那雄胺(Finasteride)是一种4-氮甾体激素化合物,是20世纪90年代美国默克公司研制开发的新一类5a-还原酶抑制剂,它能有效抑制睾酮转化成二氢睾酮,通过降低体内二氢睾酮的水平达到治疗良性前列腺增生的目的。由于非那雄胺的体内浓度较低,测定血浆中的非那雄胺需要灵敏度较高的方法,目前文献报道有HPLC-UV法、HPLC/APCI-MS—-MS法。本工作建立了一种简便灵敏的液相色谱-大气压化学电离源质谱联用(HPLC/APCI-MS)法测定
【Abstract】 To establish a method for the determination of finasteride in human plasma using HPLC-MS. The plasma was alkalized with 1 mol·L-1 sodium hydroxide and extracted with ether. The HPLC separation of finasteride was performed on a Diamonsil C18 analytical column using the mixture of methanol and water (75:25) including formic acid 1% and ammonium formate 2 mmol·L-1 as the mobile phase at a flow rate of 1.0 mL·min-1. An Agilent quadruple MSD equipped with an atmospheric pressure chemical ionization (APCI) source was used as detector and was operated in positive ion mode. Selected ion monitoring (SIM) was performed and the m/z of ions selected for quantitation were 373.5 (finasteride) and 285.2 (internal standard, diazepam). The linear range of finasteride in plasma was 1~128 ng·mL-1. The method recoveries were 98.5%~101.7%. The RSDs of intra-day and inter-day were less than 3. 3% and 7. 6%, respectively. The method is proved to be convenient, specific, accurate and sensitive enough to be applied to the pharma-cokinetic studies of finasteride in human plasma.
