Apo—AI基因多态性与鸡体组成性状的相关研究

【摘要】 载脂蛋白AI(APo—AI)是血浆高密度脂蛋白的主要成分,对胆固醇的动态平衡起到重要的作用。本研究以东北农业大学选育的肉鸡高、低腹脂系为试验材料,依据鸡Apo—AI基因的5′侧翼区序列设计引物,通过测序和PCR—SSCP方法进行SNP检测和基因型分析,探讨Apo—AI基因多态性与鸡体组威性状变化之间的关系。研究发现在Apo—AI基因序列起始密码子ATG上游163bp处存在一个A/T突变。对该突变在研究群体中进行基因型分析,结果产生3种基因型。基因型与鸡体组成性状进行的最小二乘分析结果表明BB基因型个体与AB基因型个体相比有较高的腹脂重和腹脂率(P<0.05)。表明该基因对鸡腹脂性状具有较大的影响或与控制腹脂性状的主效基因连锁。

【Abstract】 Apo-AI is a major protein component of plasma high-density lipoproteins, and plays an important role in cholesterol homeostasis. This study was designed to investigate the effect of Apo-AI gene on chicken body composition traits. The sixth generation broilers of divergent selection line for abdominal fat were used in this research. Body weight and body composition traits were measured at 7 weeks of age. Primers for 5’ flanking region in Apo-AI were designed from database of chicken genomic sequence. Polymorphisms were detected by DNA sequencing, and PCR-SSCP method was then developed to screen the population. An A/T mutation at base positions 163bp upstream of the ATG initiation codon of Apo-AI gene was found among individuals. The least square analysis showed that BB birds had significant higher(P<0. 05) abdominal fat weight and percentage of abdominal fat than AB birds. It implied that Apo-AI gene could be a candidate locus or linked to a major gene to significantly affect abdominal fat traits in chicken.
