大豆体细胞胚胎发生的组织学及转基因技术研究

【摘要】 为了建立规模化成熟化高效的植物遗传转化再生体系,保证转基因植物大量群体的获得,有利于转基因性状与其他农艺性状的综合筛选,针对具有产业化开发前景的7个优良大豆基础材料进行了体细胞胚胎发生的组织结构与转基因技术的实用性研究。利用农杆菌介导法进行了大豆未成熟子叶转化试验,通过筛选获得抗性植株。转基因植株经分子生物学鉴定为阳性,外源基因gus在转化植株中稳定表达也提供了转化成功的证据,转化频率为6.8%。转基因植株的To及T1代植株均获得了种子。应用优化的农杆菌介导转化技术体系,获得了多个品种有价值的目的基因的大豆转基因株系。
【Abstract】 For obtaining transgenic soybean, it is necessary that found the stable,simple, repeatable program which less depending plant genetypes and higher regeneraterate of tissue culture seeding. In this paper, using 7 good genetypes that could be potentialto industrialation as basic materials, the applied research of tissue culture high rateregeneration and transgenic technology are carried out, while tissue cytology structuralanalysis is to somatic embryo genesis system. Plant differentiate of the regenerate systemis observed by paraffin wax slice. Its cell splitting pattern origins from single cell, whichis excellent receptor system for gene operation. Two kinds of improved soybean tissueculture high rate regeneration systems is mediated transformation by argobacterial, andobtained resistant plant with transformation rate 6.8% by screening. Transgenic plantsare identificatated to positive by molecular biologic assay, while GUS gene expressingstably in the transformation plants is found. Using two improved high rate regeneratesystem (immature embryo and cotyledonary node ) and Agrobacterial, many valuabletransgenic strains with different target gene are obtained after mediated transformationsoybean.
