免疫荧光技术快速诊断鸡流行性腹泻的研究

【摘要】 用分离的轮状病毒与弗氏完全佐剂或弗氏不完全佐剂混匀后免疫家兔制备高免血清.分离球蛋白.用荧光素标记,建立荧光抗体快速诊断鸡流行性腹泻技术。试验结果表明.建立的荧光抗体诊断技术特异性强、敏感性高.与新城疫病毒、禽流感病毒、传染性法氏囊病病毒、大肠杆菌不发生交叉反应。人工感染试验结果表明,20日龄SPF鸡感染轮状病毒后12小时,肠粘膜深层涂片可出现较强荧光。自然感染诊断结果表明.荧光抗体诊断结果与病毒的分离鉴定一致.符合率达100%。
【Abstract】 Anti - rotavirus serum was prepared by immunised rabbits with Rotavirus and Freund’s complete and incomplete adjuvant. Then Immuno globulins were purified and labeled with fluorescein isothiocyanate. The method of rapid detection of epidemic diarrhea in chickens by immunofluorescent antibody technique was developed. The results of negative test and positive test showed that the technique had high specificity and sensitivity, and had no interaction with NDV, AIV, IBDV and E. coli. The result of artificial infection test showed that strong fluorescence appeared on intestinal mucous membrane 12 hours after 20 day -old SPF chickens were infected with rotavirus. The result of detection for natural infection showed consist ence with the result of virus isolation and identification.
