传染性法氏囊病超强毒株Gx的分离鉴定及分子生物学和抗原性分析

【摘要】 从广西某地IBD免疫后发病鸡群采取法氏囊组织。经标准化方法处理后,得到IBDV-Gx株。对Gx进行了AC-ELISA试验和致病性试验,克隆了其主要保护性抗原基因VP2.并进行了序列分析和比较。结果表明.Gx-VP2的氨基酸序列与欧洲超强毒株UK661完全相同,抗原性与欧洲超强毒株849相同,其对SPF鸡胚半数致死率(EID50)为105.25/0.2ml.以2×103EID50/只的剂量接种4周龄SPF鸡.死亡率达64%。从而证实传染性法氏囊病病毒Gx为一株符合欧洲超强毒标准和特点的国内超强毒株。

【Abstract】 The very virulent infectious bursal disease virus (vvIBDV) strain Gx was isolated from a chicken farm of Guangxi province, China, in which IBD with the mortality up to 80% broke out 5 days after immunization by serotype 1 IBD vaccine in 1996. The results of AC -ELISA, pathogenicity test, cloning and analysis of VP2 gene showed that the deduced amino acid sequence of strain Gx Vp2 was the same as vvIBDV UK661, which is now considered as the reference strain for European vvIBDVs, and the antigenicity of strain Gx was the same as a European vvIBDV strain 849. The EID50 of Gx virus was 108.25/0. 2ml; the mortality was 64% when 4 week -old SPF chickens were challenged at dosage of 2×l03EID50. It was proved that the IBDV strain Gx isolated in China was a vvIBDV according with the European standard.
