鸡肿瘤病的快速鉴别诊断技术的建立及其在广西流行病学的研究

【摘要】 本研究通过对三对引物、反应混合物及反应条件等进行优化.建立了鉴别诊断鸡最常见的三种肿瘤病即马立克氏病(MD)、禽白血病(AL)和网状内皮组织增生症(RE)的三重PCR方法;用建立的鸡肿瘤病快速鉴别诊断技术,2年来对广西139群的505羽病、死鸡的1570份组织病料进行了检测。结果MD、RE和AL的群阳性率和个体阳性率分别为87.05%与74.06%、24.46%与11.49%、14.39%与6.73%,三种肿瘤病混合感染率为1 6.83%,首次证实MD是广西鸡最主要的肿瘤病.RE和AL也有一定的流行;流行病学调查的材料还显示鸡群的免疫抑制状态、生产性能下降等与肿瘤病的发生相关。本研究的结果表明.建立的鸡肿瘤病快速鉴别诊断技术具有简便、敏感、特异、稳定的优点.不仅适用于临床鸡鸡肿瘤病的鉴别诊断.而且对肿瘤病的流行病学的调查、疫苗质量的监测以及兽医检疫也具有重要应用价值。

【Abstract】 Marek’s disease (MD), reticuloendotheliosis (RE) and avian leukosis(AL) are the most common tumors in chickens. A rapid differential diagnosis for avian tumors, which based on multiplex PCR technique was developed to amplify the specific gene fragments; 132bpr of MDV, LTRs of ALV and REV of virus genomes, the specificity of the multiplex PCR were also evaluated. The whole procedure, from sampling to the final conclusion, was finished within one day’s work-hour (5hrs. ) in our protocol.1570 tumor/suspect tumor samples of 505 chickens from 139 flocks in Guangxi were detected using the PCR -based protocol, which was developed for the rapid differential diagnosis of avian tumors. MD was the most common tumor in the survey (87. 05% of the flocks and 74. 06% of the birds detected were positive), RE was pretty common(24. 46% of the flocks and 11. 49% of the birds detected) and then was AL (14. 39% of the flocks and 8. 08% of the birds detected). Dual and triple infections were accounted for 16. 83% (85/505). Proventriculus was the most detectable organ for MD and RE while liver was the optimal organ for AL detection.The results demonstrated that the PCR protocol is a specific, sensitive and rapid tool for the differential diagnosis of avian tumors, which provided a possibility for the development of rapid differential detection kit foi avian tumors. It will play an important role in epidemiological investigation, and veterinary quarantine for a-vian tumors.
