生物气溶胶新型采样介质的研制

【摘要】 为了提高生物气溶胶的采样效率,为采样标准化奠定实验基础,研制了一种能适合大多数生物气溶胶的新型采样介质。利用大肠杆菌和粘质沙雷氏菌在静态和动态分别对该介质进行了测试,确定了4个配方后又对其进行了初步应用测试,正交试验结果表明,采样介质与对照组相比,在静态下对3种细菌的保护效率对大肠杆菌4次实验平均提高58.2%,40.5%,44.1%,20.5%,对粘质沙雷氏菌平均提高24.7%,对金黄色葡萄球菌平均提高15.4%。初步确定的4种介质配方PD1,PX2,TD1,TX2具有较好的保湿效果,4种采样介质在动态条件下对待测微生物具体保护效果(以提高百分比计)为:PD1 226%(153/47),PX2 553%(111/17),TD1 150%(120/48),TX2 268%(419/114),介质对随后的核酸检测有轻微影响,但可以通过氯仿抽提或快速核酸抽提技术消除,介质对微生物的动态保护作用明显,静态下具有一定的保护效果,正好符合介质既要提高保护效果、又不刺激微生物生长的要求。

【Abstract】 In order to standardize bioaerosol sampling and to raise sampling efficiency, a new sampling medium for detecting bioaerosols was developed. The sampling medium was tested by using Escherichia coli, Staphylococcus aureaus and Serratia marcescens strains under static and active conditions. Preliminary applications were performed using an AGI-10 and a high volume samplers. The results showed that the average recovery rates were raised to 24.7%, 58.2%, 40.5%, 44.1%, 20.5% and 15.4% respectively for six consequent experiments under the static condition and 30 min at room temperatures. Four kinds of sampling media were determined after static experiments, whose names are defined as Samplution PD1 PX2, TD, and TX2 respectively. For the active condition, the protection efficacy was as follows: PD, 226%( 153/47), PX2 553%(111/17), TD, 150%(120/48) and TX2 268%(419/114). The Samplution did have some effects on the subsequent nucleic acid detection, but can be avoid by using nucleic acid extraction procedure. The newly developed samplution can be applied to the detection of bioaerosols, it not only has certain protection to microbes in the samples, but does not facilitate the growth of microorganisms in samples.
