大鼠颈椎间盘组织基因表达谱的研究

【摘要】 目的:研究正常和退变模型大鼠颈椎间盘的基因表达谱,分析大鼠颈椎间盘基因表达水平的变化,用于指导诊断和治疗。方法:建立动静力失衡性人鼠颈椎间盘退变模型,从5月对照组和模型组人鼠颈椎间盘中抽提mRNA,经反转录分别用Cy3、Cy5荧光标记,获得两组动物椎间盘cDNA的探针,再与包含512个cDNA基因表达谱芯片杂交,结果由激光扫描仪扫描并用软件进行图像分析、标准化处理、ratio值分析、聚类分析。结果:共筛选出18条表达有差异的基因,11条基因表达量明显下降(ratio<0.4),其中细胞信号转导类有4条;7条基因表达量明显上升(ratio>2.5)。这些基因在颈椎间盘退变的表达模式与其他的报道类似。结论:退变火鼠椎间盘发生基因表达的调节变化,细胞内外的信号转导参与了椎间盘退变过程,这些研究对进一步阐明颈椎病在基因水平上的发病机理有重要的作用。
【Abstract】 Objective: To investigate the genes differentially in normal and degeneration cervical intervertebral disc of rat and some cervical-intervertebral-associated genes which can be used for diagnosing and treating . Methods: The models of rat degeneration of cervical intervertebral disc caused by imbalance between the dynamic and static forces was established. cDNA microarray chips containing 512 cDNAs were used to investigation on the gene expression pattern in cervical intervertebral disc of rat. Results: there 18 genes expressed in cervical intervertebral disc were screened out , comprising 11 up-regulated and 7 down-regulated, respectively. This genes express in cervical spondylosis is similar to other reports. Conclusion: Signal transduction pathways regulate the process of the degeneration of cervical intervertebral disc. Investigation on the gene expression pattern plays an important role in degeneration of cervical intervertebral disc pathogenesis
