玉米自交系7922幼胚愈伤组织诱导条件的优化

【摘要】 以玉米自交系7922的幼胚为外植体,探讨了4℃保存时间、激素种类及浓度、碳源种类及浓度、培养基成分、AgNO3等因素对玉米愈伤组织诱导的影响。结果表明:取4℃保存时间不超过7d的幼胚,以8114、MB、MS、N6为基本培养基,选用40g/L的蔗糖浓度或20g/L的蔗糖+10g/L的葡萄糖作碳源,加入1-2mg/L的2,4-D,是较适宜的诱导条件。AgNO3对于愈伤组织的诱导也有一定的促进作用。
【Abstract】 Factors influencing the callus induction of immature embryos in maize inbred line 7922 such as duration of immature embryos storage at 4℃, auxin types and concentration, culture medium, carbon source and concentration, were studied in this experiment. The results showed that the better conditions of callus induction were less than 7 days storage at 4℃ , 8114, MB, MS, N6 culture medium, 1. 5-2.0mg/L 2,4-D, 40g/L sucrose or 20g/L plus 10g/L glucose as carbon source. The AgNO3 also had positive effect on callus induction.
