脑瘫仔兔血清及脑组织中神经元特异性烯醇化酶含量的研究

【摘要】 目的 本研究旨在探讨神经元特异性烯醇化酶(NSE)能否作为脑瘫早期诊断生物学指标及反映各脑区损伤情况.方法 本实验通过给生后3天仔兔腹腔分次注射胆红素的方法(总量300mg/kg)制备出免脑瘫动物模型,并观察脑组织不同区域病理变化;采用酶联免疫法测定血清和脑组织中NSE含量.结果 血清中NSE含量末次注药后12小时实验组较对照组明显升高(P<0.05),而生后45天脑瘫兔与对照组比无明显差异.脑瘫兔海马、基底节区NSE含量较对照组减少(p<0 05),而脑干、小脑、皮层区NSE含量与对照组比较无显著性差异(P > 0 05).结论 血清中NSE含量可作为胆红素致脑损伤早期诊断的敏感指标;胆红素致脑瘫兔海马、基底神经节区神经元数量减少.

【Abstract】 Objectives To study if neuro-specific enolase(NSE)can be a biological marker of early diagnosis and the damage in diffent brain regions of cerebral palsy. Method The model of cerebral palsy was established by administration intraperitoneally with bilirubin(300mg/kg) in postnatal 3 days rabbits. The pathological changes of different brain reaions were observed. The levels of NSE in serum and brain tissues were measured. Results The levels of senum NSE of cerebral palsy newborn rabbits at 12 hours after last administration were significantly higher than those in the control group(P< 0. 05), but there was no difference between the levels of serum NSE in cerebral palsy newborn rabbits at 45 days after last administration and those the control. The contents of NSE in hippocampus and basal ganglia in cerebral palsy new born rabbits were reduced, but these was no change of NSE in brainstenu Cerebellum.,cortex. Conclusion The levels of serum NSE might be helpful in the early diagnosis of brain damage induced by bilirubin. The number of neuron was decreased in hippocampus,basal ganglia.
