【摘要】 衣壳蛋白VP60为免病毒性出血症病毒唯一的结构蛋白,与诱导抗病毒感染的免疫反应直接相关.本试验自感染RHDV致死的兔肝脏组织中提纯病毒,提取病毒总RNA,以RT-PCR方法扩增出全长2010bp的VP60片段.将RT-PCR产物克隆到T载体中,并对重组质粒进行酶切分析及PCR测定.结果初步表明克隆片段为完整的目的基因,从而为基因工程苗及分子诊断试剂的研制奠定了基础.更多还原

【Abstract】 Capsid protein VP60 is the only structure protein of RHDV, which can induce immune response to RHDV. In this study, RHDV were purified form liver samples of rabbits which killed by RHDV, and total RNA were repaired. A 2010 bp fragment of vp60 was amplified by RT-PCR then was cloned into pMD-T vector. The recombinant plasmid were identification by restriction enzyme digestion and PCR, the results showed elementarily that the cloning gene was the target gene. This experiment make the possibility for the farther study to new vaccine and diagnosis reagent.更多还原
