质控大小鼠肿瘤动物模型的建立及应用

【摘要】 对来自不同研究单位保存的S180、H22、EAC及L1210瘤细胞株的细胞学及生物学特性进行了对比研究,证实了由不同单位保存的同一名称的瘤细胞株的特性已存在显著差异.为了得到可进行质量控制的瘤细胞株,对以上4种瘤细胞株分别进行了克隆,对所获得克隆细胞株的特性进行了比较研究,证明从一个瘤株中所获得的不同克隆细胞株的特性存在差异,对这些克隆细胞株的细胞学特性、建立动物模型的生物学特性及治疗学反应性进行了研究,认为S2D9、H2D8、E2G8及L3E11克隆细胞,在一定程度上可以分别代表S180、H22、EAC及L21O瘤细胞株特性.用采购的及自制的核酸探针、抗体检测不同克隆细胞株,证明了这些探针及抗体可以作为检测这些克隆细胞株的试剂:克隆细胞株可以通过克隆化进行分纯,达到到长期稳定培养的目的.

【Abstract】 We compared some cytological and biological characteristics of S180, H22, EAC and L1210 cell lines from different institutes, respectively. The results testified that there were significant differences among same named cell lines that stored in different institutes. To obtain the cell strains that can be controlled on quality, we isolated the 4 different cell lines by the single cell cloning technique and compared characteristics of cloned cell strains, respectively. We showed that the different biological characteristics were existed among cloned cell strains from the same cell line. S2D9, H2D8, E2G8 and L3E11 cloned cells may be suitable when they are inoculated in inbred mice as quality control tumor animal models. We examined the differences of mRNA expression and proteins in cell lines and its different cloned cells, and found that cDNA probes and antibodies could distinguish these cloned cells.
