狐米草大规模离体培养繁殖技术研究

【摘要】 以狐米草的种子和幼穗为外植体材料,分别从盾片和颖壳部位诱导产生愈伤组织。诱导培养基中的固化剂以Gelrite或Phytagel替代Agar,种子诱导愈伤组织的频率达55%,幼穗诱导愈伤组织的频率达90%。培养基中附加椰子汁能明显减轻愈伤组织的褐化现象,有利于愈伤组织的不断增殖。选择带有绿芽点的胚性愈伤组织可长期继代增殖,在分化培养基上植株再生频率达100%,平均每块愈伤组织分化出10多株试管小植株。培养基中添加IBA和活性炭促进试管苗生根,生根率达90%以上。在苗床上间隔浇灌植物营养液和人工海水,试管苗移栽成活率高于80%。

【Abstract】 Seeds and young spikes of Spartina patens were used as explants. Calli were induced from scutellum of seed or glume of young spike. The inductiverates of callus were up to 55 % and 90 % , respectively, in seeds and young spikes on medium with solidifying agents-Gelrite or Phytagel. Coconut water could reduce the brown phenomenon of callus during callus subculture. Embryo-genic calli have maintained the plant regeneration possibility for 2 years. 10 - 20 plantlets were regenerated from each embryogenic callus. The rooting rate of plantlets was above 90% on medium supplemented with IBA and active carbon. Through irrigating the plant nutrient solution and artificial sea water, the survival rate of transplanted plants was over 80%.
