苹果白化茎段外植体的离体再生和基因转化
Study on the Shoot Organogenesis and Gene TransFormation from Etiolated Stem Explant of Apple (Mains x domestica Borkh)

【摘要】 “皇家嘎啦”苹果新梢节间外植体不定芽的再生数目,经白化处理,顶部第一节间较第二、第三、第四节间分别高2、8、73倍,比同一部位正常新梢高7倍;而且白化处理的新梢顶部第一节间外植体均有β-葡糖醛酸酶(GUS)表达区域,比常用的叶片外植体高4倍.与根癌农杆菌(Agrobacterium tumefaciens)EHA105(p35Gusint)株系共培养两周后,白化处理新梢第一节间外值体能产生9.8个表达GUS反应的愈伤组织.采用该种外植体获得了GUS基因的转基因植株.
【Abstract】 Internodal explants from etiolated ’ Royal Gala’ apple shoots were compared with those from non - etiolated shoots, for frequency of shoot organogenesis and for efficiency of β - glucuronidase (GUS) expression after cocultivation of explants with Agrobacterium tumefaciens strain EHA105 (p35SGUSint). First (youngest) internodal explants from etiolated shoots produced 2 - , 8 - and 73 - fold numbers of shoots compared to second, third, and fourth internodal explants, respectively. Moreover, these explants produced 7 - fold the number of shoots compared to similar explants from non - etiolated shoots. All first internodes from etiolated shoots exhibited GUS expressing zones and yielded 4 - fold as many GUS expressing zones compared to commonly used leaf explants from non - etiolated shoots, which exhibited GUS expressing zones in only 63% of the explants. An average of 9.8 GUS expressing calli per explant were produced on first in-temodes from etiolated shoots two weeks after cocultivation with A. tumefaciens. Transgenic plants with gusA gene were obtained and confirmed by DNA Southern Blot.
