便携式微量免疫层析色谱分析仪的开发

Development of Portable and Sensitive Immunoassay Chromatography Test Strip Reader

【摘要】 胃癌的发病率和死亡率在近年来一直居高不下,统计显示,我国每年的新病例占世界所有的42%,且胃癌的晚期治愈率不到10%,严重威胁了人们的身心健康,而胃癌的早期治愈率高达85%。研究表明,可以通过检测幽门螺旋杆菌中的尿素酶的含量来实现胃癌的早期诊断,从而降低胃癌的死亡率。免疫层析检测技术是一种高效的检测方法。由于它具有速度快、使用简便以及结果稳定等特点,从而使得它在药物残留、疾病标记物以及环境污染等方面有着广泛地应用。近年来,很多课题组都开始研究新兴的纳米材料和配套的检测设备,使得免疫层析技术从单一的定性检测发展为定量、高灵敏度的检测。本文在课题组之前的研究基础上对免疫层析分析仪进行了改进和优化,研制了一套基于CMOS传感器和平板PC的定量检测设备。该设备由硬件系统和软件系统两个部分组成。硬件系统包括图像采集模块、条码扫描模块、光源模块、供电模块以及图像处理模块。并将所有模块封装在一起,实现设备一体化。软件系统的功能包括病人信息的录入、图像采集和检测以及数据的存储和载入。对于不同类型的试纸条采用了不同的去噪方法。在计算检测结果时采用了不同的方式来实现试纸条的定量检测。随后选取了不同浓度的尿素酶试纸条样品来验证本设备的稳定性和准确性,取得了良好的效果。本分析仪体积小、便携性强且拥有优秀的续航能力,对于尿素酶的检测下限可达到5 mIU,能够用于长时间的临床诊断、社区体检以及野外检测。

【Abstract】 In recent years the morbidity and mortality of the gastric carcinoma has been stayed at a high level.According to the statistics,the number of new case in our country has been about 42% of the worldwide.The cure rate of the late stage gastric carcinoma is less than 10%,which has been a severe threat to human health.But the cure rate of the early stage gastric carcinoma reaches up to 85%.According to the research,the early diagnosis of the gastric carcinoma can be realized by detecting the concentration of urea enzyme in the helicobacter pylori,leading to reducing the mortality of the gastric carcinoma.Lateral-flow immunochromatographic assay(LFIA)is a simple and efficient method for early detection.It has rapid detection rates,cost-effective and stable results features.Hence,it has now been widely used in drug residue,biomarker of disease and environment contamination.Now many groups has been started in research of the new nanometer materials and the matched detecting devices,which would make the immunochromatographic technology from the simple qualitative detecting to become a quantitative and sensitive method.This article has been made an improvement and optimization of our group’s test strip reader.In this paper,a new kind of test-strip reader was designed based on complementary-metal-oxide-semiconductor transistor(CMOS)and tablet personal computers(PCs).This reader is composed of the hardware system and software system.The hardware system is made up of image capture module、barcode scanner module、light source module、power supply module and image processing module.We packaged all the modules and realize the integration of the reader.The functions of the software system include the input of the patient’s information、image capture and detection、save and load of the data.We used different denoising methods according to different types of the test strips.We also chose different methods to realize the quantitative detection of the strip.To evaluate the stability and accuracy of this device,different concentrations of urea enzyme samples were tested.The results were very well.This test strip reader has a light weight and can run for long durations without an AC power supply.The detection limit of the urea enzyme samples reached 5 mIU.Above all,this reader can be used for long time clinical test、community examination and outdoor detection.
