短梗五加与刺五加次生代谢物组分差异研究

【摘要】 刺五加(Acanthopanax senticosus(Rupr.et Maxim.)Harms)为五加科(Araliaceae)五加属(Acanthopanax)多年生落叶灌木植物,药用刺五加主要是五加科植物刺五加的干燥根及根茎。刺五加作为一种常见的中草药已被广泛使用,人工种植刺五加起步较晚,种植技术不够完善,利用短梗五加代替刺五加还存在一定难度。目前,对不同产地、不同生境刺五加中药用成分含量的比较研究和刺五加化学成分的研究较多,但对刺五加与短梗五加的药用次生代谢物含量的区别比较研究较少。药用植物不同组织部位往往具有不同的药用成分,且次生代谢物的含量也存在较大差别,有大量临床实验证明,药用植物不同组织部位具有不同的疗效功能。不同年限刺五加各种化合物的含量也存在一定的差异,随年限的增加一部分化合物含量增加,一部分化合物分解或转化为其他物质,还有部分化合物含量并不随年限的增加而改变。现阶段针对刺五加不同年限的研究较少,现有的部分针对刺五加年限研究的报道也只是针对刺五加中某一种或两种次生代谢物含量的研究,或某类化合物的研究,不能全面的针对特异性物质含量进行质量评价。为了深入研究:(a)不同生长年限刺五加,主要药用部位根及茎中,10种次生代谢物以及19种酚类物质的含量差异,找到刺五加最适宜的采摘年限。(b)刺五加与短梗五加不同组织部位中,以10种次生代谢产物以及19种酚类物质含量差异,讨论哪些目标化合物的提取可以使用短梗五加替代刺五加资源;本文以吉林省八家子林业局刺五加和短梗五加以及黑龙江省七台河地区三年、五年、九年刺五加为材料,以UPLCMS/MS和UPLC/Q-TOF-MS为技术手段,从以下几个方面进行研究:(1)建立同时定量检测刺五加苷E、槲皮苷、芦丁、绿原酸、齐墩果酸、山奈酚、异嗪皮啶、原儿茶酸、刺五加苷B、金丝桃苷的UPLC-MS/MS方法。(2)不同年限刺五加中次生代谢物积累情况不同,本文研究了三年、五年、九年三个年限刺五加根及茎中10种次生代谢产物及酚类物质的含量变化,找出刺五加作为药用刺五加资源的最佳提取年份为五年。(3)刺五加与短梗五加不同组织部位中10种次生代谢产物的含量及酚类物质的相对含量不同,且分布情况不同。根据次生代谢产物的不同,选择相应的组织部位作为提取材料,可以有效的提高刺五加的利用率。
【Abstract】 Acanthopanax(Acanthopanax senticosus(Rupr.et Maxim.)Harms)is the shrubs of Acanthopanax senticosus group in Araliaceae.Acanthopanax as a common Chinese herbal medicine has been widely used,artificial planting Acanthopanax are not perfect,the use of Acanthopanax brachypus Harms instead of Acanthopanax senticosus(Rupr.et Maxim.)Harms.is difficult.At present,there are many studies on the chemical constituents of Acanthopanax senticosus in different habitats and different habitats and the chemical composition of Acanthopanax senticosus,but the difference between the medicinal secondary metabolites of Acanthopanax senticosus and Artificially cultivated Acanthopanax Senticosus Comparative studies are less.The different tissues of medicinal plants often have different medicinal components,and the content of secondary metabolites is also quite different.A large number of clinical experiments have proved that different parts of medicinal plants have different therapeutic functions.There were also some differences in the contents of Acanthopanax senticosus in different years.With the increase of the number of compounds,some compounds were decomposed or decomposed into other substances,and some of the compounds did not change with the increase of the age.At present,there are few studies on the different years of Acanthopanax,and some of the existing reports on the study of the Acanthopanax epidemics are only for the study of the contents of one or two secondary metabolites in Acanthopanax,or some kind of compounds Of the study,can not be comprehensive for the specific substance content for quality evaluation,for more rational use of Acanthopanax to provide theoretical basis.In order to further study:(a)the different growth years Acanthopanax,the main medicinal parts of the root and stem,10 kinds of secondary metabolites and 19 kinds of phenolic substances content difference,find Acanthopanax most suitable picking years.(B)In 10 secondary metabolites and 19 kinds of phenols content difference,which discuss the extraction of target compounds can be used instead of Acanthopanax sessiliflorus Acanthopanax senticosus;in Jilin province forestry bureau and eight subsidiary of Acanthopanax acanthopanxsenticacosusharms and Heilongjiang area of Qitaihe province for three years,five years,nine years in Acanthopanax as materials.UPLC-MS/MS and UPLC/Q-TOF-MS Technology,the research from the following several aspects:(1)The establishment of simultaneous quantitative detection of Acanthopanax glycoside E,quercitrin,rutin,chlorogenic acid,oleanolic acid,kaempferol,isoxidene,protocatechuic acid,acanthopanax glycoside B,gold UPLC-MS/MS method of peptides.(2)Different years of secondary metabolite accumulation in Acanthopanax thorn is different,this paper studies three years,five years,nine years three years.The content changes of Acanthopanax roots and stems of 10 secondary metabolites and phenols,find out the best extraction year of Acanthopanax senticosus as medicinal Acanthopanax senticosus for five years.(3)The contents of 10 secondary metabolites and the relative contents of phenols in different tissues of Acanthopanax senticosus and acanthopanax senticosus were different,and the distribution was different.According to the secondary metabolites,selecting the appropriate tissue parts as extraction materials can effectively improve the utilization rate of Acanthopanax senticosus.
