新型纳米材料在沙丁胺醇试纸条中的应用研究

【摘要】 论文选择沙丁胺醇(SAL)作为目标物,研究了胶体金、纳米金-聚苯胺-纳米金微球(GPG)、胶体碳三种标记抗体方法,分别建立了沙丁胺醇免疫层析分析方法,并应用于动物源性食品中沙丁胺醇残留的可视化检测。建立了沙丁胺醇胶体金标记免疫层析分析方法,在最优条件下组装试纸条,将金标抗体铺在结合释放垫上的组装方式其方法检出限为2嵋L-1;将金标抗体冻干在微孔板中的组装方式其方法检出限为1鹏L-1。选择猪肉、牛肉、羊肉和猪肝四种实际样品进行添加回收实验,样品检出限为5μg L-1。合成新材料GPG,建立了沙丁胺醇GPG标记免疫层析分析方法,在最优条件下组装试纸条,将GPG标记抗体铺在结合释放垫上的组装方式方法检出限为5μgL-1;将GPG标记抗体冻干在微孔板中的组装方式其方法检出限为2 μg L-1。选择猪肉、牛肉、羊肉和猪肝四种实际样品进行添加回收实验,样品检出限为10μgL-1。SB4型号炭黑平均粒径约为50 nm。建立了沙丁胺醇胶体碳标记免疫层析分析方法,在最优条件下组装试纸条。将碳标抗体铺在结合释放垫上不可行;将碳标抗体冻干在微孔板中以及在溶液状态下与标准品混合上样两种方式灵敏度基本一致,方法检出限为2μgL-1。选择猪肉、牛肉、羊肉和猪肝四种实际样品进行添加回收实验,样品检出限为10μgL-1。三种标记材料所建立的免疫层析分析方法灵敏度基本一致,采用胶体金作为标记物灵敏度略高;对于纳米材料GPG和胶体碳,其批间差异小,稳定性优于胶体金;炭黑作为工业材料,成本远远低于氯金酸,采用胶体碳作为标记物可以节约成本,同时,胶体碳标记抗体,平均每根试纸条抗体用量约为使用胶体金作为标记材料的1/12,可以节约大量抗体,有利于实现试纸条产品的市场化。
【Abstract】 In this study, three kinds of labels:colloidal gold, colloidal carbon and nanogold-polyaniline-nanogold microspheres (GPGs) were studied and applied for immunochromatography test strip to determine Salbutamol(SAL) respectively. Conditions of the designed strips procedure were optimized.An immunochromatographic rapid test strip based on colloidal gold as label was developed for the determination of salbutamol. The detection limit of the SAL test strip with colloidal gold-labeled antibody jetted into the conjugate release pad was 2 μg L-1. The detection limit of the SAL test strip with colloidal gold-labeled antibody lyophilized on microplate was 1 μg L-1. Pork, beef, lamb and liver were used as real samples, and the detection limit of samples were 5μg L-1.A new type of nanomaterial named GPG was synthesized, and immuno-chromatographic rapid test strip based on this material has been developed. The detection limit of the test strip with GPG-labeled antibody jetted into the conjugate release pad was 5 μg L-1. The detection limit of the SAL test strip with GPG-labeled antibody lyophilized on microplate was 2 μg L-1. Pork, beef, lamb and liver were used as real samples, and the detection limit of samples were 10 μg L-1.The average particle size of SB4 carbon black was 50 nm, and immuno-chromatographic rapid test strip used colloidal carbon as a visual marker has been developed. Assemblage method with colloidal carbon-labeled antibody jetted into the conjugate release pad was infeasible. The detection limit of the test strip used two other assembly methods was all the same, which was all 2μg L-1. Pork, beef, lamb and liver were used as real samples, and the detection limit of samples were 10μg L-1.The sensitivity of test strips utilized three different kinds of nanomaterials respectively was nearly the same. For GPG and colloidal carbon, the differents between batches were small, and the stability of these two materials was much better than colloidal gold. The price of colloidal carbon was much lower than colloidal gold, what’s more, used colloidal carbon as a visualized label could save nearly 11 times antibody compared with colloidal gold. Used colloidal carbon as a label to develop an immunochromatographic rapid test strip can reduce the cost, and the productions easily scaled up.
