石榴皮多糖的制备、鉴定及其抗氧化活性研究

【摘要】 石榴(Punica granatum L.)为石榴科石榴属落叶灌木或小乔木,原产于地中海至非洲西部地区,主要分布于亚热带及温带地区,其果实、果皮、种子、根皮、花及叶均可入药,在我国主要有5个栽培区域即陕甘豫地区、新疆地区、安徽怀远地区、山东峄城地区和西南地区。目前,关于怀远石榴皮多糖的研究还未有报道。本研究探索怀远石榴皮多糖的提取条件,为怀远石榴皮多糖的开发提供理论依据。1、石榴皮多糖的制备工艺及抗氧化研究:采用水浸乙醇沉淀法从怀远石榴皮中提取多糖,通过单因素实验和正交试验,确定石榴皮多糖的最适提取条件为:料液比为1:10,提取温度为80℃,时间为2 h,提取次数为3次,多糖得率为11.13%。并采用不同的体外抗氧化实验,研究石榴皮多糖的抗氧化活性。结果显示,石榴皮多糖具有很强的还原力和清除羟自由基的活性,适度螯合亚铁离子的能力和1,1-二苯基-2-苦基肼自由基清除活性。2、响应面优化石榴皮多糖提取研究:以石榴皮为原料,以多糖提取率为指标,采用水浸法提取石榴皮多糖,提取的多糖用苯酚-硫酸光度法测定其含量,采用响应曲面法优化石榴皮多糖的提取条件进行优化,得最佳工艺条件为提取温度80℃、提取时间60 min、料液比1:15,该条件下石榴皮多糖的实际提取率可达8.66%。3、石榴皮多糖的纯化技术及其抗氧化活性研究:以怀远石榴皮为原料,以多糖得率为指标,研究提取剂乙醇浓度、大孔树脂D101、RC-NH4、阳离子交换树脂001×7对多糖纯化的影响,研究结果表明,石榴皮多糖在70%的乙醇浓度下,纯化得率最高。D101树脂对石榴皮多糖吸附量最大,且在吸附液30℃、pH为7、浓度4 mg/mL、吸附流速1.5mL/min时吸附较强。在抗氧化活性研究中表明,石榴皮多糖在不同的抗氧化体系中均表现出一定的抗氧化活性,并且抗氧化活性随着浓度的增加而增强。

【Abstract】 Pomegranate (Punica granatum L.) is deciduous shrub or small tree of pomegranate genus in pomegranate, native to the Mediterranean Sea to the West African region, mainly located in sub-tropical and temperate regions, fruit, peel, seed, the root bark, flowers and leaves can be used as medicine and and five cultivated area of Shanxi and Gansu Henan, Xinjiang region, Huaiyuan, Shandong Yicheng and southwestern region in our country. Currently, the study has yet reported on Huaiyuan granatum polysaccharides. This study explores the Huaiyuan granatum polysaccharide extracted conditions, to provide a theoretical basis for the development Huaiyuan granatum polysaccharide.1, Granatum polysaccharide preparation and antioxidant research:flooding ethanol precipitation polysaccharide extracted from Huaiyuan granatum by single factor experiment and orthogonal experiment to determine the pomegranate rind polysaccharide optimum extraction conditions:liquid ratiol:10, extraction temperature is 80℃, time of 2 h, extracted for 3 times, the polysaccharide was 11.13%. And experiments, different in vitro antioxidant research the granatum polysaccharide antioxidant activity. The results revealed that the the pomegranate rind polysaccharide has strong reducing power and clear the activity of the hydroxyl radical, moderate chelating ability of ferrous ions and 1,1 dipheny1-2-picryl hydrazine (DPPH) radical scavenging activity.2, Response surface optimization granatum polysaccharide extracted study:to granatum as raw material extraction rate indicators, polysaccharides, flooding extraction granatum polysaccharide, polysaccharide extracted with phenol-sulfuric acid spectrophotometric determination of its content, response surface methodologyoptimize granatum polysaccharide extraction conditions were optimized, was the optimum conditions for the extraction temperature of 80℃, extraction time 60min, solid to liquid ratio of 1:15, the condition pomegranate rind the actual polysaccharide extraction rate of up to 8.66%.3, Purification technology and its antioxidant activity of granatum polysaccharide:to huaiyuan granatum as raw material, the indicators polysaccharide was to study the ethanol concentration of the extractant, macroporous resin D101, IRC-NH4 cation exchange resin 001×7 the purified polysaccharide results show that of pomegranate rind polysaccharide in 70% ethanol concentration, purification rate highest. The D101 resin granatum polysaccharide adsorbed amount is maximum, and in the adsorption solution is 30℃, the pH is 7, the concentration of 4 mg/mL, adsorption flow rate 1.5 mL/min strong adsorption. Showed that the antioxidant activity of granatum polysaccharides in different antioxidant system are demonstrated antioxidant activity and antioxidant activity enhanced with increasing concentration.
