外源性硫化氢对去卵巢后骨质疏松症大鼠股骨生物力学性能的影响
The Influence of Exogenous Hydrogen Sulfide on The Biomechanics Property of The Femur in Ovariectomized Rats with Osteoporosis

【摘要】 目的:通过对骨密度、生物力学指标及一些简单的宏观物理指标测量,进一步证实H2S参与骨骼代谢的调控,并探讨H2S对于骨骼生物力学性能影响,为骨质疏松的治疗提供一种新思路。方法:将48只6月龄未孕雌性SPF级近交系SD大鼠以12只为一组,随机分为4组:假手术组(仅手术探查双侧卵巢)、手术组(手术切除双侧卵巢)、低剂量Na HS注射组(切除双侧卵巢12W后再以剂量30μmol/kg/d腹腔注射Na HS 12w)、高剂量Na HS注射组(切除双侧卵巢12W后再以剂量90μmol/kg/d腹腔注射Na HS 12w),历时共24周后,对四组大鼠左侧股骨行骨密度(BMD)检测,体重股骨湿重和股骨中点直径测量,及三点弯曲力学试验。观察并分析各组大鼠在这些指标上的差异。结果:相对单纯手术组高剂量Na HS注射组的骨密度及各项目生物力学指标均有明显改善,部分参数上能与假手术组水平接近.结论:H2S确实能影响骨骼代谢,并可增加去卵巢后骨质疏松大鼠股骨的矿物沉积,改善骨质疏松大鼠股骨的生物力学性能。
【Abstract】 Objective: To further validate the control of H2 S in bone metabolism and preliminarily studied that whether the control ability of H2 S in bone metabolism plays a significant role in the treatment of osteoporosis based on the index measuring of bone mineral density(BMD), biomechanical index and some simple physical indicators.Method: Dividing 48 non-pregnant SD rats of SPF with the age of 6 months into 4 groups with 12 rats in a group. They are control group(bilateral ovaries observation only by exploratory operation), operation group(surgical removal of bilateral ovaries), low doses of Na HS injection group(intraperitoneal injection of Na HS lasting for 12 W with the dose of 30μmolkg/d after resection of bilateral ovaries for 12w) and high doses of Na HS injection group(intraperitoneal injection of Na HS lasting for 12 W with the dose of 90μmolkg/d after resection of bilateral ovaries for 12w). Bone mineral density(BMD) of left femur in four groups of rats, weight, femoral wet weight, diameter measurement of femoral midpoint of the femur and three point bending mechanical test are conducted after lasting for a total of 24 weeks. Observe and analyze the differences in these rat indicators between groups.Result: Compared with relatively simple surgical groups, bone mineral density(BMD) and biomechanical indexes in each project in high doses of Na HS injection group have markedly improved, in which some parameters are close to those in sham-operation group.Conclusion: It is indeed that H2 S can influence the bone metabolism, and increase mineral deposition and improve the biomechanical properties in femur of ovariectomized rats with osteoporosis.
