愈创素促进大鼠创面愈合过程中作用的初步研究

【摘要】 目的:本实验通过建立大鼠皮肤创面模型,探索愈创素(又称植物愈合素)在动物创面愈合过程的作用及机制。方法:实验前一周将20只Wistar大鼠(来自吉林大学白求恩医学院动物实验中心),雄性,置于洁净动物实验室饲养,实验前一天禁食,实验时将大鼠腹腔注射10%水合氯醛(300mg/kg),待麻醉后,0.6%硫化钠脱毛,生理盐水清洗裸露区皮肤,背部脊柱旁每侧切去直径为1.8cm创面两个,每个面积2.54cm2,创面距离为2.0cm,两侧共四个创面,创面深达筋膜层,创面制作后不包扎,颈部固定防自咬套,待清醒后单笼饲养,进行常规喂食。创面制备6小时后将创面随机分成四组,实验组(涂抹愈创素),对照组1(涂抹生理盐水),对照组2(涂抹碘伏溶液),对照组3(涂抹表皮生长因子),总实验创面80个,每组各20个创面,以上各组于术后当天6小时后涂抹一次,后每天固定时间涂抹一次,直至痊愈,每天观察并测量、记录创面愈合情况。通过制备大鼠创面模型,测量和计算在创面愈合过程中瘢痕厚度、瘢痕宽度、检测创面愈合过程中顺序时间创面组织CD34和VEGF的表达情况。结果:1、创面愈合后创面瘢痕厚度和宽度分析愈合优良程度发现,生长因子组≥愈创素组>碘伏组>生理盐水组。2、愈创素组在创面肉芽中CD34及VEGF表达峰值最高,愈合过程中表达总量最多。结论:1、通过在创面愈合过程中CD34及VEGF的表达以及标本观察,发现愈创素可以加快创面愈合。2、通过对瘢痕厚度和宽度的观察发现,愈创素可能影响瘢痕的发生发展。

【Abstract】 Objective:This experiment aims to discusses the role of the healing elementin the animal wound healing process and mechanism through making rat skin wound model.Methods:A week before the experiment,put experimental animals in our clean animal laboratory, and the day before the experiment injecting10%chloral hydrate (300mg/kg), wait for after anesthesia,0.6%sodium sulphide removal, saline washing exposed areas of skin, cutting each side of the back near the spinal to a diameter of1.8cmwound, each area is2.54cm squared, wound distance of2.0cm, on both sides of a total of four wound, the wound deep fascia layer,After production don’t dress wound, the neck fixed bite the set ofprevention, making the rats stay awake in single cage, free to eat.After6hours the rats is divided into four groups randomly,the experimental group (daub guaiac element), control group1(saline) daub, control group2(daub iodine volts solution), control group3(daub the epidermal growth factor),80total experimental wound, each group20wounds, above each group on postoperative day6childhood daub, after daub a fixed time everyday, until the wound healed, measuring and recording the wound healing of every day. Thro ugh the preparation, measurement and calculation in the process ofwound healing, testing scar widths equence in wound healing timeand CD34and VEGF expression of wound tissue.Results:1.Through analysising scar thickness and width of the wound healing finedegree, found that the growth factor group was the element or group>iodine v> saline group.2.The first group of CD34and VEGF expression in the woundgranulation peak is highest, most total expressed in the healing process.Conclusion:1The element in the process of wound healing can promote woundcapillary and accelerate the healing of the wound by expressing CD34and VEGF.2Through the observation of the thickness and width of the scar, wefound that the Guaiac element may affect the development of scar.
