【摘要】 [目的]：通过前列灵胶囊对慢性前列腺炎大鼠治疗作用的实验研究,评价其治疗效果,探讨前列灵胶囊对慢性前列腺炎炎症调控网络及氧化应激水平的影响,为治疗慢性前列腺炎中药的开发应用奠定实验和理论基础。[方法]：将60只大鼠驯化喂养一周后,随机分为6组,即空白组、阳性对照组、模型组、前列灵胶囊高剂量组、中剂量组、低剂量组。每组10只,各组用25%乌拉坦1gmg量,经腹腔注射麻醉,无菌条件下取下腹正中切口,打开腹腔,提出膀胱及两侧精囊,暴露前列腺背叶。其中空白组分别经前列腺双侧背叶注入无菌注射用水O.1ml,其余各组分别注入25%消痔灵注射液0.1ml,缝合后恢复1周。给药方法：空白组和模型组每日灌胃一次,每次灌胃与实验组等剂量的生理盐水；阳性对照组(前列通瘀胶囊)：每只大鼠经灌胃给予前列通瘀胶囊混悬液剂量相当于60kg成人用量20倍；前列灵高、中、低剂量组：每组大鼠给予前列灵胶囊混悬液灌胃,剂量分别相当于60kg成人用量20倍、10倍、5倍；均每日灌胃一次；连续给药30天,每天观察并记录大鼠活动、体重及精神状态变化,于末次给药后24小时,称每只大鼠的体重,每只大鼠采股动脉血3-5m1,离心冷藏,断头处死,摘除前列腺组织,前列腺组织用4%甲醛固定液固定。采用ELISA法分析各组大鼠血清中IL-6、ACP和SOD、MDA的含量。用S-P法检测前列腺组织中GSH、GST、GSH-px含量,用电镜观察前列腺组织中亡因子Bax.Bcl-2的变化。[结果]：前列灵胶囊可降低前列腺炎大鼠血清中IL-6.ACP.MDA和升高SOD的含量,以及升高慢性前列腺炎大鼠前列腺组织中GSH. GST.GSH-px的含量,前列灵胶囊还具有上调Bax蛋白含量和降低Bcl-2蛋白含量的作用。[结论]：前列灵胶囊对大鼠慢性前列腺炎有明显的治疗作用,其作用机理与降低前列腺炎大鼠血清中IL-6、ACP、MDA和升高SOD含量有关,以及升高慢性前列腺炎大鼠前列腺组织中GSH.GST. GSH-px的含量有关,前列灵胶囊还具有上调Bax蛋白含量和降低Bcl-2蛋白含量的作用。更多还原

【Abstract】 [Objective]:To evaluate the effect of treatment that Qianlie Ling Capsules on chronic prostatitis in rats, to investigate the effect of Qianlie Ling Capsule on the regulatory network and oxidative stress level in chronic prostatitis and to provide experimental and theoretical basis for the development and application of traditional Chinese medicine for treatment of chronic prostatitis[Method]:60male rats, which were15months old and Weight250-350g, domesticating and feeding after one week,were randomly divided into6groups of10each (blank group, positive control group, model group,Qlian Lie Ling Capsule high dose group, Qlian Lie Ling Capsule medium dose group, Qlian Lie Ling Capsule low dose group), Each given25%urethane(1g/kg) anesthetized by intraperitoneal injection, open the abdominal cavity on the abdominal incision,pull out bladder and two sides of seminal vesicle, exposed the dorsal lobes of the prostate under aseptic conditions.The blank control group respectively was injected of normal saline0.1ml through bilateral dorsal lobe prostate, the other groups were injected with25%Xiaozhiling injection0.1ml, sutured tissue and recovered for1week.Method of administration:The blank group and the model group daily gavage once, saline content which were administrated equaled to the Chinese Medicine content of experiment group; The positive control group (Qianlietongyu capsule):each rat was orally given Qianlietongyu capsule suspension dose which was equivalent to20times adult dosage (weight60kg); Qian Lie Ling high, medium, low dose group:rats treated with Qian Lie Ling capsule suspension gavage, the dose which was equivalent to20times,10times,5times adult dosage (60kg), this three groups were daily gavage once; successive administration for30days, observed and recorded the weight and mental state changes in activity of rats every day,24h after the last administration, said the weight of each rat, took each rat venous blood3-5ml which was centrifuged and refrigerated, then decapitated each rat and excised prostate tissue.The excised prostate tissue was fixed with4%formaldehyde fixed liquid, ELISA method was used to analysis of the content of IL-6, ACP and SOD, MDA factors in the serum of the rats, S-P method was used to detect the changes of GSH, GST,and GSH-px factors in prostate tissue,electron microscope was used to observed the expression of apoptosis factor Bax, Bcl-2in prostate tissue.[Result]:Qian Lie Ling capsule can reduces the content of IL-6, ACP, MDA factors and increase the content of SOD in the serum of the prostatitis rats,and increase the content of GSH, GST, GSH-px factors in the prostate tissue of the prostatitis rats. Qian Lie Ling capsule also can up-regulate the content of Bax protein and down-regulate the content of Bcl-2protein. [Conclusion]:Qian Lie ling capsule has obvious therapeutic effect on chronic prostatitis rats, its mechanization relates to reduces the content of IL-6,ACP,MDA factors and increase the content of SOD factor in the serum of the prostatitis rats, and increase the content of GSH,GST,GSH-px factors in the prostate tissue of the prostatitis rats.Qian Lie ling capsule also can up-regulate the content of Bax protein and down-regulate the content of Bcl-2protein.更多还原
