P63、IKKβ在宫颈癌中的表达及意义

【摘要】 目的:检测宫颈鳞癌及其癌旁正常宫颈组织中p63和IKKβ的表达及意义。方法:1.利用Real-Time PCR方法检测21对宫颈鳞癌及其癌旁正常宫颈组织中TAp63和△Np63mRNA的表达情况。2.利用免疫组化方法检测36对宫颈鳞癌及其癌旁正常宫颈组织中P63、△Np63和IKKβ蛋白的表达情况。结果:1.宫颈鳞癌中TAp63和△Np63mRNA的相对表达量分别为0.156±0.040(x±s)和0.264±0.067(x±s),显著高于癌旁正常宫颈组织中的相对表达量,分别为0.061±0.019(x±s)和0.032±0.008(x±s),两组间差异均具有统计学意义(P<0.05)。2.宫颈鳞癌及其癌旁正常宫颈组织中△Np63mRNA相对表达量与TAp63mRNA相对表达量的比值分别为1.801±0.654(x±s)、0.576±0.173(x±s),差异具有统计学意义(P<0.05)。3.p63蛋白在宫颈鳞癌及其癌旁正常宫颈组织中的阳性表达率分别为91.7%(33/36)、33.3%(12/36),差异具有统计学意义(P<0.05)。p63蛋白在低分化鳞癌组与中分化鳞癌组的阳性表达率分别为100%(7/7)、89.7%(26/29),差异无统计学意义(P>0.05);在肿瘤直径≤4cm组与>4cm组中的阳性表达率分别为90.0%(27/30)、100%(6/6),差异无统计学意义(P>0.05);在肿瘤浸润深度<1/2肌层组与≥1/2肌层组的阳性表达率分别为89.3%(25/28)、100%(8/8),差异无统计学意义(P>0.05)。4.△Np63蛋白在宫颈鳞癌及其癌旁正常宫颈组织中阳性表达率分别为88.9%(32/36)、36.1%(13/36),差异具有统计学意义(P<0.05)。ANp63蛋白在低分化鳞癌组与中分化鳞癌组的阳性表达率分别为100%(7/7)、86.2%(25/29),差异无统计学意义(P>0.05);在肿瘤直径≤4cm组与>4cm组中的阳性表达率分别为90.0%(27/30)、82.3%(5/6),差异无统计学意义(P>0.05);在肿瘤浸润深度<1/2肌层组与≥1/2肌层组的阳性表达率分别为89.3%(25/28)、87.5%(7/8),差异无统计学意义(P>0.05)。5.IKKβ蛋白在宫颈鳞癌及其癌旁正常宫颈组织中的阳性表达率分别为100%(36/36)、50.0%(18/36),差异具有统计学意义(P<0.05)。IKKβ蛋白在低分化鳞癌组与中分化鳞癌组,肿瘤直径≤4cm组与>4cm组,浸润深度<1/2肌层组与≥1/2肌层组中的阳性表达率均为100%,各组间差异均无统计学意义(P>0.05)。6.宫颈鳞癌及其癌旁正常宫颈组织中,P63与IKKβ蛋白的表达呈正相关(r=0.591,P<0.05)。7.宫颈鳞癌及其癌旁正常宫颈组织中,△Np6β与IKKβ蛋白的表达呈正相关(r=0.492,P<0.05)。结论:1.癌旁正常宫颈组织中TAp63mRNA的表达高于△Np63mRNA,在宫颈鳞癌组织中两者表达均上调,其中△Np63mRNA表达高于TAp63mRNA,提示两者表达失衡可能参与宫颈癌的发生。2.P63、△Np63蛋白在宫颈鳞癌中均呈显著高表达,其阳性表达率均与肿瘤大小、浸润深度及病理分级无关,提示P63可能参与宫颈癌的发生。3.IKKβ蛋白在宫颈鳞癌中呈显著高表达,其阳性表达率与肿瘤大小、浸润深度及病理分级无关,提示IKKβ可能参与宫颈癌的发生。4.P63及其亚型△Np63蛋白在宫颈鳞癌及癌旁正常宫颈组织中的表达均与IKKβ蛋白的表达呈正相关,提示P63和IKKβ在宫颈癌的发生中可能具有协同作用。

【Abstract】 Purpose:To detect the expression of p63and IKKβ in squamous cell carcinoma of cervix paired with adjacent normal tissue,and to explore its significance.Methods:We measured the mRNA expression of TAp63and△Np63in a series21cervical squamous cell cancer paired with adjacent normal tissue by quantitative real-time polymerase chain reaction. The expression of P63、ANp63and IKKβ protein was detected in a variety of36cases of cervical squamous cell cancer paired with adjacent normal tissue by immunohistochemistry.Results:1. The relative level of TAp63and△Np63mRNA in the tissue of cervical squamous cell cancer was0.156±0.040and0.264±0.067, respectively, which were markedly higher than those in adjacent normal tissues was0.061±0.019and0.032±0.008, respectively,and there were significant differences between two groups (P<0.05).2. The ration of the relative level of△Np63mRNA and that of TAp63mRNA in the tissue of cervical squamous cell cancer and adjacent normal tissues was1.801±0.654and0.576±0.173, respectively,and there were significant differences between two groups (P<0.05).3. The expression rate of P63protein in the tissue of cervical squamous cell cancer and adjacent normal tissues was91.7%(33/36)and33.3%(12/36) respectively, there were significant differences between two groups (P<0.05). The expression rate of P63protein in the tissue of cervical squamous cell cancer had no obvious correlation with the tumor size、tumor invision and tumor differentiation(P>0.05).4. The expression rate of△Np63protein in the tissue of cervical squamous cell cancer and adjacent normal tissues was88.9%(32/36)and36.1%(13/36) respectively, there were significant differences between two groups (P<0.05). The expression rate of△Np63protein in the tissue of cervical squamous cell cancer had no obvious correlation with the tumor size、tumor invision and tumor differentiation (P>0.05).5. The expression rate of IKKβ protein in the tissue of cervical squamous cell cancer and para-cancer tissues was100%(36/36) and50.0%(18/36) respectively, there were significant differences between two groups (P<0.05). The expression rate of IKKβ protein in the tissue of cervical squamous cell cancer had no obvious correlation with the tumor size、tumor invision and tumor differentiation (P>0.05).6. The expression rate of P63protein was positively correlated with the expression of IKKβ protein in the tissue of cervical squamous carcinoma and para-cancer tissues(r=0.591,P<0.05).7. The expression rate of ANp63protein was positively correlated with the expression of IKKβ protein in the tissue of cervical squamous carcinoma and para-cancer tissues (r=0.492,P<0.05).Conclusion:1. The expression of△Np63mRNA was much higher than TAp63mRNA in the tissue of cervical cancer, while the former was much lower than the later in para-cancer tissues, so the unbalance between TAp63and△Np63may be potential factor to promote cervical cancer. 2. The expression of P63and△Np63protein were both markedly higher than that in para-cancers tissues,but they had no obvious correlation with the tumor size、invision and differentiation,P63perhaps associated with the occurrence of cervical cancer.3. The expression of IKKβ protein was markedly higher than that in para-cancers tissues,but it had no obvious correlation with the tumor size、invision and differentiation, it perhaps associated with the occurrence of cervical cancer.4. The expression of P63and△Np63protein were both positively correlated with IKKβ in the tissue of cervical squamous carcinoma paired with adjacent normal tissue, P63and IKKβ maybe have synergistic effect to promote cervical cancer.
