云南峨山褐煤蜡树脂化学成分研究

A Preliminary Study on Chemical Compositions of Motan Resin from Motan Wax of Yunnan Esan

【摘要】 褐煤蜡树脂是精制褐煤蜡工业生产中从褐煤粗蜡里脱除的大量副产物,目前尚无有效利用途径。为实现褐煤蜡树脂的合理开发,本实验首次采用现代化学分离方法和波谱学方法,对云南峨山产褐煤蜡树脂成分进行了深入的化学成分研究,从中分离得到24个化合物,鉴定20个,分别为:3-木栓酮(Friedenlan-3-one,1),3-羟基-木栓醇(3-Epifriedlinol,2),3-羟基-齐墩果内酯(3),31-藿烷酸(4),29-藿烷酸(5),藿烷四醇(6),24(R)-3B-羟基豆甾醇(Stigmastanol,7),24(R)-38-5-烯-羟基豆甾醇(8),邻苯二甲酸-二(3-甲基-己烷基)酯(9),邻苯二甲酸-二(7-甲基-壬烷基)酯(10),1,3-丙烷酸乙酯(11),葵烷基环己烷(12),十六烷酸甲酯(13),十二烷酸甲酯(14),庚烷酸甲酯(15),十四烷(16),十一烷(17),葵烷(18),辛烷(19),庚烷(20)。以上20个化合物均为首次从褐煤蜡树脂中分离得到的单体化合物。对云南峨山褐煤蜡树脂的药理活性做了初步探索性研究。褐煤树脂粗划段化合物对二甲苯致小鼠耳肿胀的影响实验,以二甲苯致小鼠耳廓肿胀作为急性炎症模型,考察以肿胀、渗出为主要指标。实验结果发现:褐煤蜡粗划段化合物样品P-B, P-C, P-D,A-Ⅰ A-Ⅱ组均具有一定的抗炎活性。另外对云南峨山褐煤蜡树脂血管新生的影响研究:利用鸡胚尿囊膜模型(CAM)对云南峨山褐煤蜡树脂粗提取物各极性部位进行了血管新生活性筛选。筛选结果发现:峨山褐煤蜡树脂丙酮萃取物对血管新生具有促进作用,其中P-D,A-Ⅰ,A-Ⅱ组效果较明显具有显著的促血管新生活性。

【Abstract】 Large amount of montan resin is generated as by-product during production of refined motan resin from motan wax and can not recycle. In order to ecological utilization, twenty-four comppunds were isolated from the montan resin by modern chemical separation methods, and the structures of twenty comppunds were elucidated by spectroscopic methods. They are Friedenlan-3-one (1),3-Epifriedlinol (2),3-hydroxy-oleanane (3),31-Homohopanoic acid (4),29-Homohopanoic acid (5), Bacteriohopanetetrol (6), Stigmastanol (7), Stigmasterol (8), bis-(7-methyl-nonane) phthalate (9), bis-(-methyl-hexane) phthalate (10),1.3-propane ester ethyl (11), dacane cyclohexane (12), methyl palmitate (13), methyl dodecyl (14), Methyl heptyal (15), tettadecane (16), hendecane (17), decane (18), octane (19), heptane (20). Those twenty compounds were isolated from montan resin for the first time,In addition, the pharmacological activities of YUNNAN ESAN motan resin made a preliminary exploratory research. YUNNAN ESAN motan resin on the effects of mouse models of xylene induced ear edema. The results found:montan wax from montan wax compound sample P-B, P-C, P-D, A-I, A-II group all has anti-inflammatory activity. Moreover, The study on the angiogenesis activity of YUNNAN ESAN montan wax:The activity of the extract of montan resin and the compounds isolated from it were tested with the chicken chorioallantoic membrane (CAM) neovascularisation model. The results showed that three fractions (P-D, A-Ⅰ, A-Ⅱ group) exhibited proangiogenic activity.
