杞圆膏和二黄丸抗D-半乳糖致皮肤衰老模型小鼠的实验研究

Experimental Research on the Skin Aging Mice Model Induced by D-Galactose of Qiyuan Plaster and Erhuang Capsuie

【摘要】 目的:观察杞圆膏和二黄丸对D-半乳糖致皮肤衰老模型小鼠治疗结果的观察,以及与对照组和阳性治疗组的比较,验证杞圆膏和二黄丸是否对D-半乳糖所致小鼠皮肤衰老有拮抗作用,其作用之间的差别,初步探讨其作用机理并进行比较。方法:选用3月龄清洁型雌性小鼠50只随机分为5组,每组10只,分别为空白对照组、模型组、杞圆膏组、二黄丸组、维生素E组。空白对照组每日给予生理盐水0.2ml颈背部注射,其它组每日给予D-半乳糖1000mg/kg颈背部皮下注射,连续42天,造成皮肤衰老模型。在造模21天后,开始给予治疗处理,造模同时杞圆膏组每日给予原药0.2g灌胃,二黄丸组每日给予原药0.2g灌胃,维生素E组每日给予维生素E50mg/kg灌胃,空白对照组和模型组每日给予生理盐水0.2ml灌胃,所有组均持续灌胃21天。42天后处死小鼠,取颈背部皮肤制备组织匀浆测量过氧化氢酶(CAT)、谷胱苷肽过氧化物酶(GSH-Px)、丙二醛(MDA)、羟脯氨酸(Hyp)和超氧化物歧化酶(SOD)的含量,并取颈背部皮肤进行切片染色与组织病理学形态观察,采用计算机图像分析系统对皮肤组织表皮层和真皮层厚度进行定量分析。所有数据采用统计软件SPASS10.0进行处理,以均数(?)标准差表示,组间比较采用t检验。结果:1药物空白对照组和药物模型对照组具有显著差异(P<0.05或P<0.01);2二黄丸组和杞圆膏组不同程度降低D-半乳糖所致皮肤衰老小鼠的丙二醛(MDA)含量,统计显示作用不显著(p>0.05);3二黄丸组可以使D-半乳糖所致衰老模型小鼠皮肤中的超氧化物歧化酶(SOD)活力显著增加(P<0.05),而杞圆膏作用不显著(p>0.05);4杞圆膏和二黄丸能够非常显著增加D-半乳糖诱导的亚急性衰老小鼠皮肤表皮真皮厚度(P<0.01);5杞圆膏组和二黄丸组两中药组在本次实验中无明显差异(P>0.05)。结论:1杞圆膏和二黄丸能够增加皮肤组织中抗氧化酶(SOD、GSH-PX、CAT、Hyp)活力和含量,同时还能够增加皮肤表皮真皮厚度,延缓皮肤组织中的弹力纤维和胶原纤维的减少。具有一定的抗皮肤衰老作用。

【Abstract】 Objective: To investigate anti-aging effect and mechanism of Qiyuan plaster and Erhuangcapsule on aged mice induced by D-galactose. Method: Choose 50 three months mice andrandomly divide into 5 groups: Qiyuan plaster dose group, Erhuang capsule dose group,blank control group, model control group and vitamin E group. Mice blank control groupreceived subcutaneous injection on the back of neck with physiological saline, and mice inother 4 groups received subcutaneous injection of D-galactose (1000mg-kg-1·d-1.m) for42d to establish subacute aging model.After 22d, Qiyuan plaster-treated (200mg.d-1.gastricinjection) group, Erhuang capsule-treated (200 mg.kg-1·d-1, gastric injection) group,and vitamin E-treated (50 mg·d-1, gastric injection) group, blank control group andmodel control-treated (0.2ml.d. physiological saline, gastric injection) group, To calculatethe statistical after 22 d. the dorsal skin samples were collected to determine SOD、CATactivities and MDA、GSH-PX、hydroxyproline contents, and to calculate and analyze thedepth of surface tissue and dermis tissue from the back neck of mice withphotographs-analyzing system of computer. Result: 1. There was a significant differencebetween blank contol groups and model control group (p<0.05 or p<0.01). 2. The erhuangcapsule group and the Qiyuan plaster group can reduce the MDA content induced byD-galactose in mice’s the blood, but the effect was not significant. 3. The erhuang capsulegroup can obviously enhance senile model induced by D-galactose mice’s blood serumSOD activeness (P<0.01), but compares with the other control groups, the Qiyuancapsule group’s the effect was not significant. 4. The erhuang capsule and the Qiyuanplaster can obviously enhance the depth of epithelium and dermis of sub-senile mice’smodel induced by D-galactose (P<0.01). 5. There was no significant difference betweenthe erhuang capsule group and the Qiyuan plaster group (p>0.05). Conelusion: Qiyuanplaster and Erhuang capsule has good anti-aging effects on the aged mice, which isprobably due to effects of increasing the content and activist of epithelium and dennistissue, and the influence of increasing depth of the epithelium and dermis tissue, displaying the progressing of degeneration of elastic fiber and collagen fiber of the tissues and organsto drop.
