右美托咪定对神经病理性痛大鼠脊髓背根神经节p-JNK表达的影响

【摘要】 目的:观察右美托咪定对坐骨神经慢性压迫性损伤(CCI)所致神经病理性疼痛大鼠脊髓背根神经节p-JNK表达的影响,探讨右美托咪定镇痛的作用机制,为右美托咪定的临床应用提供实验基础和理论依据。方法:雄性SD大鼠54只,体重180-220g,采用随机数字表法,将大鼠随机分为:假手术组(S组)、CCI组(C组)和右美托咪定组(D组),每组18只。S组大鼠仅分离坐骨神经不结扎,C组和D组建立CCI模型,D组于CCI术后即刻开始至术前一天,每天腹腔注射右美托咪定50ug/kg,S组和C组注射等容量生理盐水。于术前1d,术后3、7、14d每组随机取6只大鼠测定机械痛阈(MWT)和热痛阈(TWL)。测定痛阈后,处死大鼠,取损伤侧L4-6脊髓背根神经节,免疫组化法检测脊髓背根神经节p-JNK的表达。结果:痛阂的变化:各组TWL、MWT基础值无差别。与S组比较,C组和D组术后TWL、MWT明显降低(P<0.05);与C组比较,D组术后MWT及TWL升高(P<0.05);与术后7d时比较,C组和D组于术后3、14d时MWT和TWL升高(P<0.05)。脊髓背根神经节p-JNK阳性神经元表达变化:与S组比较,C组和D组术后脊髓背根神经节p-JNK的表达上调(P<0.05);与C组比较,D组术后脊髓背根神经节p-JNK的表达下调(P<0.05);与术后7d时比较,C组和D组于术后3、14d时p-JNK表达下调(P<0.05)。结论:在本实验中,右美托咪定能有效的减轻CCI大鼠的痛温觉过敏,并维持一定的时间。脊髓背根神经节p-JNK参与了神经病理性痛的信号转导过程,右美托咪定可抑制p-JNK的表达减轻大鼠慢性神经病理性痛。

【Abstract】 Objective:To observe the effects of dexmedetomidine on sciatic nerve chronic constriction injury (CCI) induced expression of neuropathic pain in rat spinal cord dorsal root ganglion p-JNK, to explore the mechanisms of dexmedetomidine analgesia, to provide experimental and theoretical basis for the clinical application of dexmedetomidine.Methods:54male SD rats, weighing180-220g, were randomly, the rats were randomly divided into:sham operation group (S group), CCI group (group C) and dexmedetomidine group (D group),18rats in each group.The rats in group S were only isolated sciatic nerve without ligation, C group and D group, model CCI, D in CCI group after operation until the day before surgery, a daily intraperitoneal injection of dexmedetomidine50ug/kg, S group and C group were injected with saline. On the preoperative Id, after3,7,14d randomly selected from each group6rats determination of mechanical pain threshold (MWT) and heat pain threshold (TWL).Determination of threshold, the rats were killed, the dorsal root ganglia lateral L4-6spinal cord injury,immunohistochemistry was used to detect the expression of p-JNK in spinal dorsal root ganglion.Results:The changes of pain threshold in each group:TWL, MWT basis of indifference. Compared with S group, C group and D group after TWL, MWT decreased significantly (P<0.05);compared with C group, D group after MWT and TWL increased (P<0.05); compared with postoperative7d, Cgroup and D group after3,14d MWT and TWL increased(P<0.05).Expression of p-JNK in dorsal root ganglion neurons:compared with S group, the upregulation of the expression of p-JNK in dorsal root ganglion and spinal cord of C group and D group after surgery (P<0.05); compared with C group, D group after p-JNK expression in dorsal root ganglia spinal cord cut (P<0.05); compared with postoperative7d, C group and D group after3,14d down regulated expression of p-JNK (P<0.05).Conclusion:In this experiment, effects of dexmedetomidine hydrochloridecan effectively reduce the CCI rat nociceptive hypersensitivity, and maintain a certain amount of time.Dorsal root ganglion p-JNK is involved in the signal transduction of neuropathic pain dexmedetomidine can inhibit the expression of p-JNK reduces chronic neuropathic pain in rats.
