根皮苷对高脂血症仓鼠降血脂作用的研究

【摘要】 根皮苷属于植物黄酮类中的二氢查尔酮苷,具有多种生物活性。研究表明,根皮苷具有防治糖尿病、抗氧化作用和改善记忆的能力。本文以仓鼠为动物模型,主要对根皮苷的降血脂作用和其对高脂血症仓鼠的抗氧化作用以及其降血脂机理进行研究。本文采用高效液相色谱法对样品根皮苷的纯度进行了测定,色谱柱为C18柱,流动相为乙腈和0.1%的磷酸水溶液进行梯度洗脱,检测波长为280nm。测得样品根皮苷的纯度为75.02%。通过高脂血症仓鼠模型的建立以及不同剂量的根皮苷对高脂血症仓鼠的喂饲,对根皮苷的降血脂作用及机理进行研究。在膳食中添加0.3%-0.9%根皮苷可以显著降低高脂血症仓鼠血清中总胆固醇(TC)、甘油三酯(TG)和非高密度脂蛋白胆固醇(non-HDL-C)的含量,显著升高高密度脂蛋白胆固醇(HDL-C)的含量,同时也显著降低血脂水平综合指标TG/HDL-C和动脉硬化指数(AI);剂量组与模型组比较,肝脏中TC和TG含量均显著降低。这表明根皮苷具有降血脂效果。经过对仓鼠抗氧化指标的测定,结果表明根皮苷可以显著降低高脂血症仓鼠血清、肝脏、心脏、肾脏中脂质过氧化物MDA的生成;剂量组与模型组比较,仓鼠血清、肝脏、心脏和肾脏中的SOD活力、CAT活力、GSH-PX活力、T-AOC均有明显的升高,只有血清T-AOC没有显著性,其余均差异显著。因此说明了根皮苷具有很好的抗氧化性能。根皮苷可以显著降低高脂仓鼠的血糖含量,剂量组与模型组相比,胆碱酯酶(CHE)活力也明显的增强,说明根皮苷在一定程度上可以减少肝损伤。本文通过对仓鼠肝组织胆固醇代谢相关酶(如HMG-CoA还原酶、LDL受体、胆固醇-7α-羟化酶)mRNA表达水平的测定,初步对根皮苷的降血脂机理进行探讨。经上述实验证明,根皮苷具有显著的降血脂和抗氧化作用。
【Abstract】 Phloridzin is a dihydro-chalcone glycoside in plant flavonoids, with a variety of biological activity. Studies have shown that phloridzin may prevent and treat diabetes, have antioxidant ability and improve memory. In this paper, hamsters as the research model,hypolipidemic effects, antioxidant of hyperlipidemic hamsters and hypolipidemic mechanism of phloridzin was mainly studied.In this paper, we determined purity of the sample phloridzin by high-performance liquid chromatography method. The column was C18column. A binary H2O-0.1%H3PO4(solvent A) and acetonitrile(solvent B) gradients was used for elution.Detection wavelength was280nm. It was showed that the purity of sample phloridzin was75.02%.By set the model of hyperlipidemia hamsters and fed in hyperlipidemia hamsters different doses of phloridzin, we studied the effect and mechanism of phloridzin hypolipidemic.0.3%-0.9%phloridzin adding dietary may significantly reduce the value of total cholesterol(TC), triglyceride(TG) and non-high density lipoprotein cholesterol(non-HDL-C) in serum, increase high density lipoprotein cholesterol(HDL-C) in serum, and also reduce the overall target TG/HDL-C and atherogenic index(AI) of lipid levels. Dose groups compared with model group, the liver TC and TG levels were significantly reduced. This indicates that phloridzin has hypolipidemic effect.Antioxidant indicators for the determination of hamsters, the results show that phloridzin can significantly reduce the MDA formation of lipid peroxide in serum, liver, heart and kidney of hyperlipidemia hamsters. Dose groups compared with model group, SOD activity, CAT activity, GSH-PX activity and T-AOC in serum, livers, hearts and kidneys of hyperlipidemia hamsters were increased, only T-AOC in serum is not significant, the rest were significant differently. Therefore it shows that phloridzin has good antioxidant resistance.Phloridzin can significantly reduce blood glucose levels of hyperlipidemia hamsters. Dose groups compared with model group, Cholinesterase(CHE) activity was significantly enhanced, it shows that phloridzin can reduce the liver injury to a certain extent.By measuring the enzymes of hamster liver mRA expression related to cholesterol metabolism, such as HMG-CoA reductase, LDL-R and CYP7A1, hypolipidemic mechanism of phloridzin was studied. The results of experiments showed that phloridzin possess hypolipidemic activity and antioxidant activity.
